DESCRIPTION:As part of the seminars organized by the Biological and Medical Sciences Department (SBM) at Bordeaux University\, Prof. Pierre-Emmanuel Rautou has been invited by Clotilde Billottet (Team 03 – Liver Cancers and Tumor Invasion). \nPierre-Emmanuel Rautou\, Center for Inflammation Research\, UMR 1149 Inserm\, Paris \nFriday\, December 15\, 11 a.m.\, BBS amphitheatre\, 2 rue Dr Hoffmann Martinot Bordeaux \n“Les vésicules extracellulaires\, des biomarqueurs et vecteurs d’information dans la cirrhose” \nProf. Pierre-Emmanuel Rautou (MD\, PhD) is a clinical specialist in Hepatology with a PhD in vascular biology.  He is Professor of Hepatology at the University of Paris and Hôpital Beaujon (Clichy\, France) since 2016. He is the head of the splanchnic hemodynamic laboratory at Beaujon Hospital (Clichy\, France) since 2012. He is also leading an INSERM team at the Paris Research Center on Inflammation (INSERM Unit 1149) since 2019. His research is dedicated to the study of the role of vessels in liver diseases. On the clinical side\, he is interested in primary vascular liver diseases (Budd-Chiari syndrome\, extrahepatic portal vein thrombosis and obstruction of intrahepatic portal veins)\, as well as in vascular involvement in cirrhosis development and complications. On the basic side\, his objectives are to identify the role of extracellular vesicles and of autophagy in liver diseases.      \nAbstract :\nExtracellular vesicles are membrane-bound vesicles containing proteins\, lipids\, RNAs and microRNAs. They can originate from both healthy and stressed cells\, and provide a snapshot of the cell of origin in physiological and pathological circumstances. Various processes that may give rise to the release of extracellular vesicles occur in liver diseases\, including hepatocyte apoptosis\, hepatic stellate cell activation\, liver innate immune system activation\, systemic inflammation\, and organelle dysfunction (mitochondrial dysfunction and endoplasmic reticulum stress). Numerous studies have therefore investigated the potential role of extracellular vesicles as biomarkers in liver diseases and particularly in cirrhosis. But extracellular vesicles are also vectors of information between cells and organs in the setting of cirrhosis. This lecture will cover both aspects.

DESCRIPTION:Mélanie Moreau\, team 03 – Liver Cancers and Tumor Invasion\, will present her thesis on Friday\, December 8\, 2:30 p.m. in the CGFB amphitheatre\, 146 rue Leo Saignat\, Bordeaux. \nsupervisor: Frédéric Saltel \n  \nInvolvement of invadosomes in tumor invasion through two examples\, Ruvbl2 in molecular organization and DDR2 in angiogenesis \nMetastasis is the main cause of death in cancer patients. Around 90% of all deaths are attributed to tumor invasion. Invadosomes are F-actin structures known to be involved in the metastatic process\, particularly through their function of extracellular matrix degradation. This promotes the migration of cells into the extracellular matrix\, and enables them to cross anatomical barriers such as vessel walls. In this thesis\, I focused on the role of Ruvbl2 in invadosome formation and activity. Ruvbl2 is an AAA+ATPase\, member of the R2TP complex\, which plays a chaperoning role in the formation of big protein complexes and is found enriched in rosette-shaped invadosomes. I have contributed to demonstrating that Ruvbl2 is involved in the process of invadosome formation and\, more specifically\, in the organization of these structures into rosettes\, via the R2TP complex. This complex protects several proteins involved in invadosome formation\, including Fak\, from autophagic degradation. I was also interested in the regulation of angiogenesis via invadosome formation and activity in endothelial cells. I demonstrated that\, in the presence of type I fibrillar collagen\, proteasome interacts with DDR2 receptor and negatively regulates invadosome activity. Proteasome inhibition drastically increases invadosome degradation activity. This inhibition stimulates DDR2 activation\, mainly by Src. These results underline the importance of a balance between Src and proteasome activity to avoid cell death. Finally\, this work provides a better understanding of mechanisms of induction\, formation and regulation of invadosomes. It highlights a set of proteins and functions linked to these invasive structures that could be pharmacologically targeted to inhibit metastasis.     \n  \nKey words : Cancer – Invadosomes – Invasion – Ruvbl2 – R2TP – DDR2

DESCRIPTION:Ce symposium sur les cancers pancréatiques est organisé par des scientifiques l’équipe 2 et de l’équipe 8 (Samuel Amintas\, Sandrine Dabernat\, Serge Evrard\, Majid Khatib\, Christophe Laurent\, Véronique Vendrely\, Simon Pernot). \nt will take place from 16 to 18 October 2023 at the Domaine du Haut Carré in Talence. \nInformation and registration : https://www.pcs-afrcp.com/ \nDeadline for submission of abstracts and early birds registration : 06-09-2023 \nAlthough pancreatic ductal adenocarcinoma (PDAC) represents less than 2% of new cancers\, it is constantly increasing (14\,000 new cases/year) and is one of the main causes of cancer-related mortality. Indeed\, the vast majority of patients present at diagnosis with locally advanced or metastatic disease. Its incidence nearly equaling its mortality rate underscores the incurable nature of this cancer. Despite the progress made in understanding the pathogenesis of PDAC\, and the improvement of treatment regimens\, the survival rate after treatment remains poor. Thus\, basic and clinical research remain the best allies of patients and medical teams to make significant breakthroughs and improve the prognosis of this dreaded disease. \nThe ‘Pancreatic Cancer symposium 2023’ will be the third edition of an international meeting that brings together researchers and clinician-scientists at the forefront of knowledge of pancreatic cancer. It is organized by a voluntary group of French researchers (AFRCP). Considering recent progress\, this event will focus on current and future themes in the field. In particular\, the themes of the following sessions\, including interdisciplinarity potentially leading to new advances\, will be proposed to the international research community: identification and targeting of new tumor vulnerabilities\, exploration of the desmoplastic reaction and the tumor microenvironment\, new modalities and technologies for pancreatic cancer research\, knowledge and understanding of early and metastatic disease. \nGiven the international nature of the congress\, we wish to interact with the European association DICE\, https://digestivecancers.eu/ \, in a final session to make a European inventory of patient involvement in the construction of research.

DESCRIPTION:The CRCT postdocs association was created in 2022 by Benoît Aliaga\, Chloé Bessière and Steffen Fuchs. It gathers the 31 postdocs who are currently present in our research center. DESCRIPTION:Séminaire de Robert Bristow : “Evolutionary Trajectory of Aggressive Prostate Cancer: Composite Role of Genomics and Microenvironment” \nRobert Bristow\, Manchester Cancer Research Centre\, will speak on February 28 at 2:00 p.m. in the IBGC conference room\, invited by Majid Khatib (team 2). \n\nRobert Bristow is the Director of Manchester Cancer Research Centre and Director of CRUK Major Centre. He was previously a Clinician-Scientist and Professor within the Departments of Radiation Oncology and Medical Biophysics at the University of Toronto and a Senior Scientist at the Princess Margaret Cancer Centre. His primary research interests are in tumour hypoxia\, DNA damage signalling and DNA repair in tumours\, and the genomics of prostate cancer progression and cancer treatment response. He is particularly interested in novel clinical trials that intensify cancer therapy to prostate cancer patients whose tumours harbour aggressive genetic changes and hypoxic sub-regions. He has over 300 published papers and book chapters. He is twice a Canadian Foundation for Innovation (CFI) awardee. DESCRIPTION:Dr Barak Rotblat seminar:”How do cells survive glucose starvation” \nBarak Rotblat\, Ben Gurion University (BGU)\, Israël is invited by Andreas Bikfalvi (team 01) for a webinar on February 14 at 11am. \nAbstract: Cells use glucose as a primary source of energy and have developed ways to detect changes in glucose levels to activate survival mechanisms. One such mechanism is the inhibition of a protein called mTOR\, which is responsible for controlling cell growth. It was previously thought that this inhibition leads to a reduction in protein synthesis and preservation of ATP\, but our research has found that the true mechanism is actually a decrease in the translation of mRNA that codes for ACC1\, an enzyme involved in fatty acid synthesis. This leads to a decrease in fatty acid synthesis\, NADPH consumption\, and ROS levels. This mechanism is conserved in human. mouse and yeast cells and exploited by tumor cells to generate tumors in the brain\, a low-glucose environment. \nProf. Rotblat studies the molecular biology of cancer by examining the role of long non-coding RNAs and regulators of mRNA translation. Among other functions\, long non-coding RNAs (lncRNA) control regulation in cells. Protein content analysis shows that a cancerous cell is very similar to a normal cell from the same tissue; for example\, a cancerous breast cell is more similar to a breast cell than to a cancerous cell in the lung. However\, while the protein profiles of healthy and cancerous cells from a given tissue are very similar\, their lncRNA content is very different. Therefore\, lncRNA represents a promising drug and therapeutic target. Regulating mRNA translation is key in the cellular mechanism by which tumor cells adapt to stress. Prof. Rotblat and his colleagues seek drug targets and inhibitor molecules aimed at processes that are used by cancer cells to adapt to starvation conditions\, such as found in a tumor. \nZoom ID 873 524 180 13

DESCRIPTION:We are pleased to announce that on September 29-30\, we will be holding the first conference of our new institute\, the BRIC Kick-off meeting. \nThis first event is intended to celebrate the creation of the BRIC and to initiate a series of future symposiums and other congresses within the BRIC. \nThis kick-off meeting will be organized over a day and a half and will include the presentation of the scientific projects of the BRIC members but also of 4 renowned guests: 3 people from SAB Jean-Ehrland Ricci (C3M\, Université Côte d’Azur\, France)\, Ruth Scherz-­Shouval (Weizmann institute of Science\, Israel)\, Urszula Hibner (IGMM\, France) and Phil Jones (The Wellcome Sanger Institute\, Royaume Uni). Finally\, the second day will close with a round table discussion between patient associations and researchers on the theme of patient/researcher collaboration. \n11 sessions are reserved for presentations by BRIC researchers. We already invite each of the team leaders to communicate to us the person who will represent their team during this meeting via this internal link. DESCRIPTION:Anthony Bouter\, UMR5248 CBMN\, Bordeaux and Frédéric Saltel\, U1312 BRIC\, Bordeaux will speak at the Oncosphere’s interdisciplinary seminars the 7 septembre at 12pm\,conference room at IBGC and videoconference. \n \nFrédéric Saltel\nBRIC INSERM 1312\, Team 03 – Liver Cancers and Tumor Invasion. \n\n\nCancer cells transfer invasive properties through tracks\, a novel collagen-associated extracellular vesicle entity \nMetastasis is the leading cause of cancer-related deaths. During this process\, tumor cells acquire invasive and migratory capacities in order to invade the surrounding tissues. To achieve this\, the tumor microenvironment including the extracellular matrix (ECM) is altered to facilitate cancer cell proliferation and dissemination. Extracellular vesicles (EVs)\, such as exosomes or migrasomes\, are already known to induce pro-tumor features such as migration\, promoting tumor development and metastasis formation. Here we highlight a new type of EVs\, referred as tracks\, released by cancer cells during migration and specifically attached along collagen fibers. These tracks\, identified by discoidin receptor 1 (DDR1) enrichment\, are promoted when cell-ECM interactions are increased\, such as in tumors. We characterized these tracks\, their ultrastructure as well as their molecular composition in terms of proteins and nucleic acids\, showing that they are different from classical EVs known so far. Moreover\, these tracks are very stable structures and can be internalized by surrounding cells. After internalization\, they modify the differentiation status and the phenotype of recipient cells\, promoting epithelial to mesenchymal transition\, matrix degradation and invasion. \nThus\, we identified a new class of collagen-associated EVs\, tracks\, that play a role in cell-cell communication by transferring invasive properties. DESCRIPTION:Alina Chan\, Broad Institute of MIT & Harvard\, will speak on July 4th at 4pm by videoconference. \nInvited by Andreas Bikfalvi\, team leader of Team 01 – Tumor and vascular biology laboratory. \n“The Origines of COVID-19” \nDr. Alina Chan is a co-author of the book\, VIRAL: The Search for the Origin of Covid-19\, and a researcher with a background in medical genetics\, synthetic biology\, and vector engineering. At the Broad Institute of MIT & Harvard\, Dr. Chan is currently creating next generation vectors for human gene therapy. During the COVID-19 pandemic\, Dr. Chan began to investigate problems relevant to finding the origins of the SARS-CoV-2 virus and in parallel spearheaded the development of the COVID-19 CoV Genetics (covidcg.org) browser for scientists worldwide to rapidly track virus lineages and mutations by locations and date ranges of interest. Her talk will focus on what we know about the origins of COVID. The seminar should be very interesting for everyone.

DESCRIPTION:Sabine Colnot\, Centre de Recherche des Cordeliers\, Paris\, will speak on June 28 at 3:00 pm\, amphitheater 11\, Carreire campus. \nInvited by Violaine Moreau\, team leader of theTeam 03 – Liver Cancers and Tumor Invasion. \n“CRISPR-Cas9-engineered mice developing liver cancers with beta-catenin mutation: towards a better understanding of liver carcinogenesis” \nOne third of hepatocellular carcinoma (HCC) have mutations activating beta-catenin signaling. We definitely showed that these mutations are oncogenic in mice (Loesch et al.\, J Hepatol 2022)\, and are responsible for the development of HCC and mesenchymal hepatoblastoma. This mouse model and a previous one obtained through a loss of function of Apc are valuable preclinical models to better understand why a large subset of beta-catenin activated HCCs are resistant to immunotherapies. Our work further supports the hypothesis that they could be treated based on their metabolism (Gougelet et al.\, Gastroenterol 2019; Senni et al.\, Gut 2019). Lastly\, the CRISPR-Cas9 strategy appears to be an easy-to-use approach to investigate the oncogenic function of specific mutations in HCC.
