Some of the known markers not only identify the tumor cells but also contribute to cellular behavior. DESCRIPTION:As part of the Soirées Santé program\, Bordeaux University Hospital and Librairie Mollat are organizing an evening on the theme of infertility: the environment in question? \nGuests:  \nLucie Chansel-Debordeaux\, reproductive biologist\, head of the Reproductive Biology Department – CECOS and MCU-PH team 8 Biotherapies Genetics and Oncology (BioGO)  in our BRIC Unit – BoRdeaux Institut of Oncology (INSERM – Université de Bordeaux). \nFleur Delva\, public health doctor\, coordinator of the ARTEMIS center\, an environmental health prevention platform  dedicated to reproduction\, and researcher in the EPICENE team at Bordeaux Public Health  (INSERM – Université de Bordeaux). \n\n\nInfertility directly affects over three million people in France\, a figure that is rising every year. The rise in infertility is primarily due to the fact that women are now giving birth to their first child an average of 5 years later than forty years ago. It can also be linked to medical causes such as endometriosis in women or genetic abnormalities in men.   So the origin of infertility can be multifactorial\, and more and more scientific data suggests an environmental effect.\n\nOccupational and non-occupational environmental exposure to chemical substances is recognized as being able to interfere with the entire reproductive process (fertility disorders\, pregnancy pathologies\, developmental abnormalities). Numerous studies describe a link between exposure to certain families of chemical substances – endocrine disruptors – and fertility disorders. Exposure to these substances may have direct effects on gametes\, but evidence also suggests longer-term effects\, increasing the risk of chronic disease in childhood and adulthood (the developmental origins of health and disease “DOHAD” hypothesis).  What’s more\, throughout their lives\, individuals are exposed to multiple environmental factors that can have an impact on their health and that of future generations. The term “exposome” refers to the totality of an individual’s exposures over the course of a lifetime. DESCRIPTION:AFIS\, Association Française pour l’Information Scientifique\, is organizing an online conference with Professor Emeritus Jacques Robert on Tuesday March 26 at 8pm. \nAll information and registration at this link. \nCancers have been a particular source of inspiration for charlatans\, naturopaths and crudivores. Cancers carry a strong emotional charge\, are seen as a curse\, and carcinophobia leads people who are well-informed and thoughtful to turn to solutions that defy common sense and reason. If “alternative” methods of preventing and curing cancer proved as effective as they are\, the disease would have been eradicated a long time ago. And yet\, each year sees its share of charlatans appear on the juicy market of alternative “medicines” and prosper\, who with a miracle diet\, who with a new “revolutionary” treatment. Quackery is certainly dangerous on an individual level\, and famous names such as Steve McQueen and Steve Jobs have paid the price. But it is also dangerous on a collective level\, as it insidiously spreads false ideas and vain hopes throughout the population\, thus contributing to the spread of pernicious sectarian phenomena. During this webconference\, Jacques Robert will take stock of these charlatanisms and the dangers they represent. Jacques Robert is Professor Emeritus of Oncology at the University of Bordeaux since September 2019. In this capacity\, he is a member of Team 2 (Reprogramming of tumor activity and the associated microenvironment – Rytme) and an honorary hospital practitioner at the Institut Bergonié\, the Bordeaux Cancer Center\, where he headed the biochemistry laboratory for over 35 years.

Moffitt from Harvard Medical School and Dieter Henrik Heiland from the Medical Center of the University of Friborg will give a lecture. \nThis event is organized by Majid Khatib\, head of team 02 – Reprogramming of tumor activity and the associated microenvironment (Rhytme) of our BRIC unit and Thomas Daubon of the IBCG\, in collaboration with the department’s Seminars GT. \n📆 Tuesday March 26 at 2 p.m. \n📌amphitheater of the BBS building\, Carreire campus\, 2 rue Dr Hoffmann Martinot Bordeaux. \n\nSpatial transcriptomics is an innovative method that allows mapping gene expression at a spatial scale in complex biological tissues. Unlike traditional techniques which measure gene expression from homogenized tissue extracts\, spatial transcriptomics makes it possible to preserve information on the cellular localization of messenger RNAs. This emerging field offers significant advantages for understanding pathologies. By mapping gene expression within tissues\, researchers can identify specific cell types involved in disease processes\, understand the interactions between these cells\, and reveal the molecular alterations associated with diseases. Spatial transcriptomics offers a new perspective for studying pathologies by allowing detailed visualization of gene expression inside tissues\, which paves the way for a better understanding of disease mechanisms and more targeted therapeutic strategies.    \n In this context\, Majid Khatib (BRIC) and Thomas Daubon (IBGC)\, the organizers\, invited Jeffrey R. In this study\, we employed preclinical models to elucidate transcriptomic signatures associated with pancreatic cancer progression and treatment response. Through integrative analysis of gene expression profiles\, we identified key molecular pathways and biomarkers implicated in disease pathogenesis and therapeutic resistance. Furthermore\, utilizing patient-derived xenografts and organoid cultures\, we validated the clinical relevance of our findings and assessed the efficacy of targeted therapies in a personalized manner. Our results underscore the potential of transcriptomic signatures derived from preclinical models to guide the development of personalized treatment strategies for pancreatic cancer patients\, ultimately improving clinical outcomes and advancing precision oncology.      \n——————————————————————————————————————————————– \nNelson Dusetti is an oncology researcher dedicated to advancing personalized treatments for pancreatic cancer. As Co-Lead of the Department and Team Lead for Translational and Therapeutic Targets Research in Pancreatic Cancer\, his work is focused on developing innovative approaches to address the challenges of individualized chemotherapy regimens. His research emphasizes the importance of predictive biomarkers in guiding treatment decisions\, particularly in the context of limited druggable targets in advanced pancreatic cancer tumors. Dusetti’s recent study highlights the potential of comprehensive transcriptomic analysis to predict chemotherapy responses\, providing valuable insights into treatment outcomes. By integrating microenvironmental factors and employing AI-based predictive tools Dusetti and his team have made significant strides in optimizing treatment selection and improving patient outcomes. His ongoing efforts are now directed towards refining these approaches for seamless integration into routine clinical practice\, ultimately ensuring the effective transfer of their studies to the everyday care of pancreatic cancer patients. Additionally\, Dusetti is a Co-Founder and CSO of Predicting Med\, a pioneering startup dedicated to commercializing molecular tests for chemotherapy in pancreatic ductal adenocarcinoma.

DESCRIPTION:For the first edition of the prestige seminars organized by our BRIC unit\, we are delighted to welcome Dr Olivier Delattre\, Research Director at Curie Institute\, head of the Unit U830 “Cancer\, Heterogeneity\, Instability and Plasticity” (CHIP) and “Grand prix INSERM 2022” \nWe will take advantage of his visit to organize meetings with researchers and students from our unit. \nSeminar at 5pm on Tuesday February 6\, 2024\, BBS conference room\, 2 rue Dr Hoffmann Martinot. \nDr. Olivier Delattre is MD-PhD\, trained in pediatric oncology. He is Research Director at Inserm and heads the Research Unit U830 “Cancer\, Heterogeneity\, Instability and Plasticity” (CHIP) and the research team “Diversity and plasticity of children tumours” (DePiCT) at the Institut Curie (Paris). He has also created the Somatic Genetics Unit\, aimed to transfer research results to the clinic\, and heads the “Genetic and Biology of Cancer” department at the Institut Curie. He also leads the SIREDO Center (Care\, Innovation\, Research in Oncology of Children\, Adolescent and Young Adults) at the Institut Curie\, gathering clinicians and researchers to accelerate the development of new therapeutic strategies for pediatric cancers. His lab identified some key genetic alterations in Ewing sarcoma\, rhabdoid tumors\, neuroblastoma as well as in various sarcomas\, and also the Neurofibromatosis type 2 (NF2) tumour suppressor gene. His lab investigates the functional consequences of these alterations in tumour development\, and has established cellular and animal models. His current fields of interest are the study of genetic alterations associated with pediatric cancers\, the study of pediatric cancers cells-of-origin and the mechanisms of cancer cells phenotypic plasticity. Dr Delattre has published more than 400 papers\, many of them in the most prestigious journals such as Nature\, Nature Genetics\, Nature Medicine\, Cancer Cell\, Molecular Cell\, New England Journal of Medicine…\, with an H-index of 102. He received numerous awards including the “Association pour la Recherche sur le Cancer (ARC)-Leopold Griffuel” award in 2016\, the “Grand Prix de la Recherche Inserm” in 2022\, and the “American Association for Cancer Research (AACR)-St. Baldrick’s Foundation” Award for Outstanding Achievement in Pediatric Cancer Research in 2023. He is an elected member of EMBO (European Molecular Biology Organization).           \n🎤 Ewing sarcoma: From genetics to epigenetics and immunology. \nEwing sarcoma is a bone cancer mostly observed in children and young adults. The EWSR1::FLI1 gene fusion (Nature 1992) is a key driver of this malignancy ( J. Clin. Oncol.  1996\, 2010; Cancer Discov. 2014). It completely reprograms the Ewing cell-of-origin (Cancer Cell 2007; Cancer Res. 2021) through direct binding at GGAA microsatellites and subsequent transformation of these sequences into bona fide enhancers (PLoS One 2009; Nature Genetics 2015; Nature Medicine 2017; Nat Rev Dis Primers 2018). Activation of GGAA microsatellites also leads to specific transcription of intergenic regions (Cancer Cell 2021\, 2022; Mol Cell\, 2022; Cell Rep. 2022). He is Professor of Hepatology at the University of Paris and Hôpital Beaujon (Clichy\, France) since 2016. He is the head of the splanchnic hemodynamic laboratory at Beaujon Hospital (Clichy\, France) since 2012. He is also leading an INSERM team at the Paris Research Center on Inflammation (INSERM Unit 1149) since 2019. His research is dedicated to the study of the role of vessels in liver diseases. On the basic side\, his objectives are to identify the role of extracellular vesicles and of autophagy in liver diseases.      \nAbstract :\nExtracellular vesicles are membrane-bound vesicles containing proteins\, lipids\, RNAs and microRNAs. They can originate from both healthy and stressed cells\, and provide a snapshot of the cell of origin in physiological and pathological circumstances. Various processes that may give rise to the release of extracellular vesicles occur in liver diseases\, including hepatocyte apoptosis\, hepatic stellate cell activation\, liver innate immune system activation\, systemic inflammation\, and organelle dysfunction (mitochondrial dysfunction and endoplasmic reticulum stress). Numerous studies have therefore investigated the potential role of extracellular vesicles as biomarkers in liver diseases and particularly in cirrhosis. But extracellular vesicles are also vectors of information between cells and organs in the setting of cirrhosis. This lecture will cover both aspects.

DESCRIPTION:Mélanie Moreau\, team 03 – Liver Cancers and Tumor Invasion\, will present her thesis on Friday\, December 8\, 2:30 p.m. in the CGFB amphitheatre\, 146 rue Leo Saignat\, Bordeaux. \nsupervisor: Frédéric Saltel \n  \nInvolvement of invadosomes in tumor invasion through two examples\, Ruvbl2 in molecular organization and DDR2 in angiogenesis \nMetastasis is the main cause of death in cancer patients. Around 90% of all deaths are attributed to tumor invasion. Invadosomes are F-actin structures known to be involved in the metastatic process\, particularly through their function of extracellular matrix degradation. This promotes the migration of cells into the extracellular matrix\, and enables them to cross anatomical barriers such as vessel walls. In this thesis\, I focused on the role of Ruvbl2 in invadosome formation and activity. Ruvbl2 is an AAA+ATPase\, member of the R2TP complex\, which plays a chaperoning role in the formation of big protein complexes and is found enriched in rosette-shaped invadosomes. I have contributed to demonstrating that Ruvbl2 is involved in the process of invadosome formation and\, more specifically\, in the organization of these structures into rosettes\, via the R2TP complex. This complex protects several proteins involved in invadosome formation\, including Fak\, from autophagic degradation. I was also interested in the regulation of angiogenesis via invadosome formation and activity in endothelial cells. I demonstrated that\, in the presence of type I fibrillar collagen\, proteasome interacts with DDR2 receptor and negatively regulates invadosome activity. Proteasome inhibition drastically increases invadosome degradation activity. This inhibition stimulates DDR2 activation\, mainly by Src. These results underline the importance of a balance between Src and proteasome activity to avoid cell death. Finally\, this work provides a better understanding of mechanisms of induction\, formation and regulation of invadosomes. It highlights a set of proteins and functions linked to these invasive structures that could be pharmacologically targeted to inhibit metastasis.     \n  \nKey words : Cancer – Invadosomes – Invasion – Ruvbl2 – R2TP – DDR2
