Tapas Kundu to present a seminar. \nPr Tapas Kundu \nTranscription and Disease Laboratory\, Molecular Biology and Genetics Unit\, Jawaharlal Nehru Centre for Advanced Scientific Research (JNCASR) – India \n  \nChromatin meets autophagy: A tale of nonhistone chromatin protein mediated genome organization and transcription regulation \nThe eukaryotic genome is intricately organized on a beads-on-string structure into nucleoprotein filaments. These filamentous structures consist of nucleosome beads\, composed of approximately 147 base pairs of DNA and four octameric core histones. The folding pattern of these nucleosomes varies depending on the cellular state\, ensuring the overall stability and balance within the organism. In addition to DNA and histones\, various non-histone chromatin-associated proteins along with RNA molecules also play crucial roles in fine-tuning the organization and function of the genome. Our laboratory discovered that a highly abundant nuclear protein\, human transcription coactivator PC4\, is a non-histone component involved in the genome organization\, stability\, and maintenance of the epigenetic state. By employing atomic force and electron microscopy\, we have demonstrated that PC4 directly interacts with core histones and linker histone H1 thus compacting the genome very efficiently. Our experimental results obtained from ATAC-Seq and RNA-Seq analysis provide valuable insights into the regulatory role of PC4 in genome organization. We observed that when PC4 is absent\, there is a significant increase in genome decompaction accompanied by hyper-acetylation levels and abnormal gene expression patterns. Interestingly\, we discovered that embryonic development was impaired in mice lacking PC4\, indicating its essential role for survival. However\, intriguingly\, cell lines with reduced levels of PC4 exhibited enhanced cell proliferation rates and demonstrated resistance to radiation exposure. Further investigation revealed that the mechanism behind this radiation resistance involves the upregulation of autophagy pathways upon depletion of PC4. Remarkably\, our ATAC-Seq data unveiled pervasive genomic decompaction coupled with heightened acetylation specifically at multiple loci related to autophagy-associated genes within these knockdown cells. A similar occurrence was observed in both brain and spleen-specific knockout models of PC4 in mice as well as C. elegans. However\, it is worth noting that the brain-specific knockout of PC4 did not exhibit a pronounced phenotype apart from noticeable impairments in memory extinction. Interestingly\, when PC4 was absent\, there was a significant increase in the expression level of another non-histone protein that interacts with PC4. He joined JNCASR as in 1999. Prof. Kundu has made significant contributions in the area of regulation of gene expression and its link to disease and therapeutics. He is not only elucidating the mechanisms of transcription regulation through the epigenetic modifications but also targeting them to design new generation diagnostics\, as well as therapeutics. Over the years\, he has published several research papers in many international journals. Several patent applications from the laboratory have been granted and some are under process\, which include several academically important research reagents with potential commercial values\, some of which have already been commercialized by renowned companies. He is a fellow of four major national academies of India and served as an editorial board member of the Journal of Biological Chemistry (JBC) from 2011-2016. Presently\, he is serving as an “Associate Editor” of the Journal of Biochemistry (Oxford University Press)\, Series Editor of Subcellular Biochemistry (Springer Nature)\, and the reviewing Editor of eLife Journal. As a Director of CSIR-CDRI\, Lucknow\, Prof. Kundu initiated many relevant academic and drug development programs. Under his leadership\, several drug molecules reached Phase-I clinical trials and could be licensed to industries for further development. He also led and completed Phase-III clinical trials for anti-Covid 19 therapy which is awaiting DCGI approval. He was instrumental to establish the ‘Asian Forum for Chromatin and Chromosome Biology and ‘Chemical Biology Society\, India’. Besides fundamental research\, Prof. Kundu is also involved in teaching and organizing science outreach programs. The intestinal type of GC is initiated by the pathogen Helicobacter pylori\, which infects half of the world’s population. The three biogenic polyamines\, putrescine\, spermidine\, and spermine play key functions in a variety of cellular processes and are essential for cellular homeostasis. Using animal models of H. pylori-mediated diseases\, we showed that the enzyme spermine oxidase (SMOX)\, which back-converts spermine to spermidine\, plays a major role in development of gastritis and gastric dysplasia. This occurs notably through the generation of acrolein\, a reactive aldehyde that initiates lipid peroxidation and further generation of bifunctional electrophiles. A potent scavenger of these molecules attenuates gastric carcinogenesis. These models\, which include organoids\, spheroids\, organs-on-a-chip and bioprinted tissues\, are currently undergoing exponential development\, thanks in particular to the coincidence between advances in stem cell biology and technologies enabling the topology and composition of the tissue microenvironment to be controlled. As well as being an alternative to animal models\, these models are of major interest for understanding developmental mechanisms\, disease modeling\, drug screening\, regenerative medicine and personalized medicine. This research is intrinsically interdisciplinary\, requiring the collaboration of biologists\, physicians\, physicists\, chemists and engineers.    \nThe aim of this first edition is to bring together people already using or wishing to develop these models\, to share their current or future projects. During this presentation I will feature published and unpublished data indicating how transcriptional and epigenetic identities are conserved throughout cell division and differentiation. Mitotic entry correlates with the condensation of the chromosomes\, changes in histone modifications\, exclusion of transcription factors from DNA\, and the broad downregulation of transcription. We found that preventing one chromosome to condense during mitosis causes it to fail resetting transcription. Rather\, in the following interphase\, the affected chromosome contains unusually high levels of the transcription machinery\, resulting in abnormally high gene expression levels\, including various transcription factors. This subsequently causes the activation of inducible transcriptional programs in trans\, even in the absence of the relevant stimuli. Thus\, mitotic chromosome condensation exerts stringent control on interphase gene expression to ensure the maintenance of cell identity across cell divisions (1). We used adipogenesis as a model to find that SUMOylation of chromatin-bound proteins ensures the precision of gene expression and transcription factor dynamics during differentiation (2). However\, the molecular mechanisms by which SUMOylation orchestrates the rewiring of transcriptional programs during initiation of differentiation remains to be established. Our unpublished data indicate that transient\, pharmacological inhibition of SUMOylation in human preadipocytes is sufficient to prime and activate specific enhancers and transcription factors. This causes irreversible hypertranscription of adipogenic genes\, leading to excessive fat accumulation. Given that epigenetic and transcriptional regulators are major SUMO targets\, we propose that SUMOylation maintains a chromatin landscape in undifferentiated cells\, which is critical to preserving the epigenetic fate of mature adipocytes\, protecting against unrestricted adipogenic transcription. This implies that the inherent adipogenic capacity of human adipose stem cells may be tied to SUMOylation and suggests an epigenetic basis for the inter-individual variability in fat accumulation. Using adipogenesis as a paradigm to uncover new molecular pathways by which SUMOylation sustains epigenetic fates\, we expect to contribute to a broader understanding of how transitory disruptions in epigenetic regulation induce stable homeostatic shifts and lead to disease\, like obesity and cancer (3).  \n\nRamos-Alonso\, L. et al. My research in the group of Jean-Marc Egly\, at the IGBMC\, was focused on revealing the origin of gene expression defects in patients with rare genetic disorders like Xeroderma pigmentosum (1). I then moved to Norway at the Dept. of Microbiology at Oslo Univ. Hospital. For the next eight years\, I continued my research in the group of JM. Enserink on gene regulation in the model organism Saccharomyces cerevisiae. The first four years were dedicated to revealing mechanisms that synchronize cell-cycle progression and gene expression (2). In the next four years I strated investigating the mechanisms regulated by the Small Ubiquitin-like Modifier SUMO at the chromatin level during adaptive response to stress (3) and started exploring the perpetuation of epigenetic and transcriptional memory throw-out mitosis (4). Next\, I started my group at Oslo Univ. Hospital and was appointed Associate Professor at the University of Oslo\, where I have been working since 2020. SUMMARY:HDR defense of François Moisan
DESCRIPTION:François Moisan\, researcher in Team 05 – Translational research in cutaneous oncology and rare skin diseases (TRIO2) will present his HDR (Habilitation to Supervise Research) on Wednesday June 5 at 2pm\, BBS amphitheater. \nSubject: Cellular and molecular mechanisms of the antitumor response to propranolol in vascular tumors. \nFrançois Moisan works on cutaneous vascular tumors and their therapy. The serendipity discovery of the effectiveness of propranolol on infantile hemangiomas (IH) in the team in 2008 led him\, upon his return from the USA in 2013\, to work on this mechanism of action. This work revealed that (1) IH is a tumor rich in norepinephrine (NE)\, one of the natural ligands of the beta-adrenergic pathway\, the expected target of treatment. (2) The cells making up the tumor (endothelial cells and pericytes) are capable of producing and secreting NE. (3) Anti-angiogenic effect is obtained through a competitive inhibition of NE by propranolol in vitro. He was also able to demonstrate (4) a third cell type which is key in the pathogenesis and response to propranolol: the telocyte. This stromal cell involved in cellular communication\, architecture and tissue remodeling\, including in the skin\, is revealed here (5) to overexpress aquaporin-1 modulated by propranolol. The development of in vitro coculture models relevant to this study makes it possible to explore the cellular and molecular mechanisms involved in the growth and regression of hemangiomas\, with the aim of identifying new therapeutic pathways for tumors and vascular abnormalities. The group he formed within the oncodermatology team (TRIO2) is also interested in angiomas associated with GNAQ mutations and angiosarcoma-type cancers.         \n

A cornerstone of this approach is the use of high-throughput genomic profiling technologies for a deep characterization of the molecular features of tumor samples. The diversity and complexity of molecular read-outs\, combined with a rapidly evolving landscape of clinically actionable targets\, poses significant requirements for advanced data interpretation tools. In this talk\, I will present molecular data interpretation frameworks that we are actively developing to address the N-of-1 analytical challenge in precision oncology\, that is to highlight treatment opportunities for each complex cancer case in light of existing knowledge and large-scale reference datasets. Herein\, I will describe challenges and solutions with respect to molecular data integration and harmonization\, the adoption of standards for evaluating the clinical significance of genomic aberrations\, as well as the identification of relevant biomarker-guided clinical trials.     \nBio \nSigve Nakken works as a scientist at the Institute for Cancer Research at Oslo University Hospital\, Norway\, with affiliation to the Centre for Cancer Cell Reprogramming (CanCell)\, a Centre of Excellence at the University of Oslo. In addition\, he holds an Adjunct Professor position at the Department of Informatics\, University of Oslo. Nakken’s main interests are tool development for clinical interpretation of cancer genomes\, mechanisms and impact of DNA variation in cancer (soma and germline)\, and knowledge integration for precision cancer medicine. He has a background from computer science (MSc) and computational biology (PhD)\, and extensive experience with analysis of data from high-throughput sequencing technologies. He is the lead developer of multiple bioinformatics tools used in high-throughput cancer biology.

The rise in infertility is primarily due to the fact that women are now giving birth to their first child an average of 5 years later than forty years ago. It can also be linked to medical causes such as endometriosis in women or genetic abnormalities in men.   So the origin of infertility can be multifactorial\, and more and more scientific data suggests an environmental effect.\n\nOccupational and non-occupational environmental exposure to chemical substances is recognized as being able to interfere with the entire reproductive process (fertility disorders\, pregnancy pathologies\, developmental abnormalities). Numerous studies describe a link between exposure to certain families of chemical substances – endocrine disruptors – and fertility disorders. Exposure to these substances may have direct effects on gametes\, but evidence also suggests longer-term effects\, increasing the risk of chronic disease in childhood and adulthood (the developmental origins of health and disease “DOHAD” hypothesis).  What’s more\, throughout their lives\, individuals are exposed to multiple environmental factors that can have an impact on their health and that of future generations. The term “exposome” refers to the totality of an individual’s exposures over the course of a lifetime. DESCRIPTION:AFIS\, Association Française pour l’Information Scientifique\, is organizing an online conference with Professor Emeritus Jacques Robert on Tuesday March 26 at 8pm. \nAll information and registration at this link. \nCancers have been a particular source of inspiration for charlatans\, naturopaths and crudivores. Cancers carry a strong emotional charge\, are seen as a curse\, and carcinophobia leads people who are well-informed and thoughtful to turn to solutions that defy common sense and reason. If “alternative” methods of preventing and curing cancer proved as effective as they are\, the disease would have been eradicated a long time ago. And yet\, each year sees its share of charlatans appear on the juicy market of alternative “medicines” and prosper\, who with a miracle diet\, who with a new “revolutionary” treatment. Quackery is certainly dangerous on an individual level\, and famous names such as Steve McQueen and Steve Jobs have paid the price. But it is also dangerous on a collective level\, as it insidiously spreads false ideas and vain hopes throughout the population\, thus contributing to the spread of pernicious sectarian phenomena. During this webconference\, Jacques Robert will take stock of these charlatanisms and the dangers they represent. Jacques Robert is Professor Emeritus of Oncology at the University of Bordeaux since September 2019. In this capacity\, he is a member of Team 2 (Reprogramming of tumor activity and the associated microenvironment – Rytme) and an honorary hospital practitioner at the Institut Bergonié\, the Bordeaux Cancer Center\, where he headed the biochemistry laboratory for over 35 years.

Moffitt from Harvard Medical School and Dieter Henrik Heiland from the Medical Center of the University of Friborg will give a lecture. \nThis event is organized by Majid Khatib\, head of team 02 – Reprogramming of tumor activity and the associated microenvironment (Rhytme) of our BRIC unit and Thomas Daubon of the IBCG\, in collaboration with the department’s Seminars GT. \n📆 Tuesday March 26 at 2 p.m. \n📌amphitheater of the BBS building\, Carreire campus\, 2 rue Dr Hoffmann Martinot Bordeaux. \n\nSpatial transcriptomics is an innovative method that allows mapping gene expression at a spatial scale in complex biological tissues. Unlike traditional techniques which measure gene expression from homogenized tissue extracts\, spatial transcriptomics makes it possible to preserve information on the cellular localization of messenger RNAs. This emerging field offers significant advantages for understanding pathologies. By mapping gene expression within tissues\, researchers can identify specific cell types involved in disease processes\, understand the interactions between these cells\, and reveal the molecular alterations associated with diseases. Spatial transcriptomics offers a new perspective for studying pathologies by allowing detailed visualization of gene expression inside tissues\, which paves the way for a better understanding of disease mechanisms and more targeted therapeutic strategies.    \n In this context\, Majid Khatib (BRIC) and Thomas Daubon (IBGC)\, the organizers\, invited Jeffrey R. Moffitt (Harvard Medical School) and Dieter Henrik Heiland (Medical Center University of Freiburg). DESCRIPTION:Nelson Dusetti : “Developing Transcriptomic Signatures through Preclinical Models for Personalized Pancreatic Cancer Treatment” \nNelson Dusetti\, Cancer Research Center of Marseille (CRCM) is invited by Majid Khatib (BRIC) for a seminar on March 19 in the IBGC conference room. \nPancreatic cancer remains one of the most aggressive and lethal malignancies\, necessitating the exploration of personalized treatment strategies. Transcriptomic analysis has emerged as a powerful tool for understanding the molecular intricacies of pancreatic cancer and tailoring therapeutic interventions. In this study\, we employed preclinical models to elucidate transcriptomic signatures associated with pancreatic cancer progression and treatment response. Through integrative analysis of gene expression profiles\, we identified key molecular pathways and biomarkers implicated in disease pathogenesis and therapeutic resistance. Furthermore\, utilizing patient-derived xenografts and organoid cultures\, we validated the clinical relevance of our findings and assessed the efficacy of targeted therapies in a personalized manner. Our results underscore the potential of transcriptomic signatures derived from preclinical models to guide the development of personalized treatment strategies for pancreatic cancer patients\, ultimately improving clinical outcomes and advancing precision oncology.      \n——————————————————————————————————————————————– \nNelson Dusetti is an oncology researcher dedicated to advancing personalized treatments for pancreatic cancer. As Co-Lead of the Department and Team Lead for Translational and Therapeutic Targets Research in Pancreatic Cancer\, his work is focused on developing innovative approaches to address the challenges of individualized chemotherapy regimens. His research emphasizes the importance of predictive biomarkers in guiding treatment decisions\, particularly in the context of limited druggable targets in advanced pancreatic cancer tumors. Dusetti’s recent study highlights the potential of comprehensive transcriptomic analysis to predict chemotherapy responses\, providing valuable insights into treatment outcomes. By integrating microenvironmental factors and employing AI-based predictive tools Dusetti and his team have made significant strides in optimizing treatment selection and improving patient outcomes. His ongoing efforts are now directed towards refining these approaches for seamless integration into routine clinical practice\, ultimately ensuring the effective transfer of their studies to the everyday care of pancreatic cancer patients. Additionally\, Dusetti is a Co-Founder and CSO of Predicting Med\, a pioneering startup dedicated to commercializing molecular tests for chemotherapy in pancreatic ductal adenocarcinoma.
