Duran\nSpanish National Research Council (CSIC) – Séville\nGroup Leader of the Group of Metabolism and Cell Signaling\n \n“Metabolic imbalance as a therapeutic strategy in cancer.” \n  \nAbstract \nDuring last years\, a great amount of effort has been dedicated to elucidate the crosstalk between cellular metabolic and bioenergetic flows with signaling processes\, and how this interaction contributes to coordinate the growth and homeostasis of cells and tissues. Particularly\, how these interaction mechanisms are deregulated in cancer at the molecular and cellular level is a matter of intense research\, with the aim of using this knowledge to propose new therapeutic alternatives. Our research has established the processes of interaction between the metabolism of the amino acid glutamine\, the most abundant in human blood and the most important from an energetic point of view for cells\, with cell signaling via the mTOR pathway\, a protein complex essential in the regulation of cell growth and metabolism\, and autophagy. More recently\, we focused our research on metabolic adaptation associated to therapy resistance in glioblastoma (GBM)\, one of the tumor types with the poorest survival outcomes. This work resulted in the identification of FGFR1 pathway as a major regulator of the signaling and metabolic rewiring associated with temozolomide (TMZ) resistance in GBM. Hence\, we described a mechanism of resistance that operates at two major levels. First\, a p53-mediated regulation of cell cycle inducing cell cycle arrest to allow DNA repair in response to TMZ. And second\, a complete metabolic rewiring promoting lipid catabolism and preventing lipid peroxidation. Both the p53-mediated response and the metabolic adaptation are controlled by FGFR1\, as inhibition of the FGFR1 pathway completely abolishes this signaling and metabolic reprograming\, restoring sensitivity to TMZ. Our results also indicated a correlation of FGFR1 activity with poor prognosis in GBM patients\, and validated the treatment of TMZ in combination with FGFR1 inhibitors as an efficient strategy to induce tumor cell death in pre-clinical animal models. This data position the receptor FGFR1 as a very promising candidate for evaluation in future clinical approaches to limit the development of therapy resistance to TMZ in GBM patients. \n  \nBiosketch \nI obtained my PhD in Biochemistry at the University of Seville (Spain) in 2005 working on cellular bioenergetics. Then\, I joined the lab of Prof. Eyal Gottlieb at the Beatson Institute for Cancer Research (Glasgow\, UK) as a postdoctoral researcher to extend my expertise on broader aspects of cancer metabolism. Later\, I moved to the lab of Prof. Michael N. Hall at the Biozentrum (Switzerland) as a senior postdoc to study the de-regulation of cellular signaling during cancer. In 2013 I joined the Institut Européen de Chimie et Biologie (Pessac\, France) as a Group Leader (INSERM\, CR position)\, integrating my previous expertise to investigate the crosstalk between metabolism and cell signaling in cancer cells. In 2018 I moved my lab to the Centro Andaluz de Biología Molecular y Medicina Regenerativa – CABIMER\, upon my assignment as Senior Research Associate (Científico Titular) at CSIC. Since them\, I am the Group Leader of the Group of Metabolism and Cell Signaling\, and in the period 2019-2022 I was the Director of the Department of Dynamics and Cell Signaling at CABIMER. In the period 2020-2024 I was the Deputy Director of CABIMER. Currently\, I am a member of the Executive Committee of the CONEXION CANCER-CSIC\, and I serve as Associate Editor of the journal Cancer & Metabolism. Since January 2025 I serve as permanent evaluator (Gestor) of the Spanish Agency of Investigation (AEI)\, in the Biomedicine – Cancer commission.

DESCRIPTION:Marion Bouchecareilh et Samuel Amintas from Team 8 “Genetic Biotherapies and Oncology” (BioGO) invite two speakers from Paris Cité University\, Center for Inflammation Research\, INSERM 1149\, Sinkus Team (From micro to macro in cancer development) for this seminar open to the scientific community. \n👉 Aurélie Beaufrère \nMorphological and molecular heterogeneity of hepatocellular carcinoma. \nAbstract:\nHepatocellular carcinoma is the most common primary malignant liver tumor. This tumor is highly heterogeneous\, both morphologically\, with eight histological subtypes described\, and molecularly\, with distinct transcriptomic groups described that are linked to both genomic alterations and morphological aspects. Some of these characteristics impact both prognosis and response to treatment.   \nBiography:\nCurrently an MCU-PH in the Pathology Department at Beaujon Hospital\, my hospital and research work focuses on liver pathology\, particularly liver tumors. I am developing projects on the morphological and molecular heterogeneity of primary liver tumors\, as well as multimodal artificial intelligence projects integrating pathological\, radiological\, and molecular data.  \n  \n👉 Etienne Becht \nHigh-throughput single-cell quantification of 100s of proteins using conventional flow cytometry and machine learning. \nAbstract: \nTechniques enabling single-cell resolution proteomic analyses are limited in terms of the number of cells analyzed and/or the number of proteins measured. To overcome these limitations\, we have developed “Infinity Flow\,” a method for analyzing flow cytometry data in parallel using machine learning models. By enabling the pseudo-quantification of hundreds of proteins in millions of single cells\, this technique allows for in-depth mapping of the microenvironment of complex tissues. We are currently applying it to the study of liver cirrhosis and hepatocellular carcinoma.    \nBiography: \nAs an engineer by training\, I completed a thesis in cancer immunology under the joint supervision of Hervé Fridman (PUPH) and Aurélien de Reyniès (bioinformatician) at the Cordeliers Research Center in Paris. There\, I developed MCP-counter\, a method for estimating the composition of the tumor microenvironment from the bulk transcriptome. I then completed two postdoctoral fellowships\, first at the Singapore Immunology Network on mass cytometry data analysis\, then in Seattle on parallelized flow cytometry data analysis. I then worked in R&D at Servier (Computational Medicine team) before joining Valérie Paradis’ laboratory at the hospital as an INSERM research fellow. My project focuses on profiling the hepatic microenvironment using parallelized flow cytometry and developing bioinformatics methods for analyzing cytometry data.     \n

DESCRIPTION:What is the purpose of a CRYO-FIB-SEM in Biology and Biomaterials in Bordeaux? (Présentation en français)\nEtienne Gontier\, PhD.\nResponsable BIC – Electronique\nIngénieur de recherche HC\nBordeaux Imaging Center\n\n\nAbstract:\n\nA cryoFIBSEM is a high-resolution scanning electron microscope equipped with a focused ion beam (FIB) column and a cryo-observation stage. In addition to being a conventional SEM\, samples can be imaged in 3D to obtain high-resolution volumes (3nm voxel) after abrasion using the FIB beam\, with or without correlative microscopy. It is also possible to generate these volumes from vitrified samples and capture the 3D ultrastructure in a state close to its native state. In addition\, soft materials such as polymers\, biological samples\, and many other systems can deform\, melt\, or become amorphous under FIB milling and SEM imaging conditions at room temperature. Thanks to cryo-FIB/SEM\, the sample is cooled to cryogenic temperatures (-130°C) in order to minimize these artifacts. This allows FIB cross-sections and SEM images to be produced with +/- chemical analysis of beam-sensitive materials\, with minimal deformation. Finally\, this technology enables\, among other things\, the preparation of cryogenic FIB lamellae for cryo-MET analysis.\n\nThe aim of this presentation will be to introduce this comprehensive\, innovative new electron microscopy equipment\, which is new to Bordeaux and the surrounding region. Thus\, initially\, the context in which this purchase is being made and the technological aspects will be explained. Next\, and most importantly\, the various possible applications will be presented in the form of concrete examples. We will conclude with a discussion.

DESCRIPTION:On the occasion of Léa Pechtimaldjian‘s thesis\, Team 5 (TRIO2) invites Bernhard Wehrle-Haller\, member of the jury\, to present a seminar to the scientific community. \nBernhard Wehrle-Haller PhD\nUniversity Medical Center\, Geneva\, Switzerland \n“The multiple roles of integrins in the assembly of fibronectin fibrils in the tumor stroma” \nAbstract:\nThe formation and growth of tumors depends on oxygen supply\, but also on mechanical support through increased synthesis of the extracellular matrix. Increased extracellular matrix stiffness enhances the survival capacity of tumor cells under therapeutic stress. At the same time\, tumor cells stimulate host cells to produce a supportive extracellular matrix. The mechanisms of this feedback are not well understood\, and there is no treatment to reverse the fibrotic response in the tumor stroma. Since integrins are essential receptors for tumor binding to the extracellular matrix\, but are also involved in the synthesis of extracellular matrix fibers\, we asked about the nature of intracellular connections and intracellular signaling that control how integrins are used by tumor or stromal cells in tumors. DESCRIPTION:We invite Kevin Boyé to give a seminar open to the scientific community. \nDr. Kevin Boyé\nCRCN INSERM Paris Cardiovascular Research Center – 56 Rue Leblanc\, 75015 Paris\nCo-founder of D2B3 – 101 College Street\, 06511 New Haven\, CT\, USA \n \n“Unlocking the Blood–Brain Barrier during Glioblastoma progression” \n📆 Tuesday\, January 27\, 2 p.m.\n🚩 BBS Conference Room\, 2 rue Dr Hoffmann Martinot\, Carreire site\, University of Bordeaux \nAbstract \nBlood vessels in the brain form 600 kilometers of specialized blood-brain barrier (BBB) that protect the brain from toxins and pathogens\, thereby ensuring homeostasis and proper neurological function. However\, BBB impermeability also severely limits brain drug delivery during many neurological disorders. \nGlioblastoma (GBM) is the most aggressive primary brain tumor with a median survival of 12–15 months. Although BBB integrity is partially disrupted within the tumor core\, the highly invasive nature of GBM cells into the BBB-competent brain parenchyma strongly limits the delivery and efficacy of therapeutic molecules\, leading to inevitable recurrence after surgical resection. Hence\, developing tailored therapeutic strategies that can transiently and safely modulate BBB permeability without compromising its protective function remains a major unmet clinical need. \nWe recently identified the endothelial receptor Unc5B and its ligand Netrin-1 as central regulators of BBB integrity. Single-cell RNA sequencing revealed that this pathway controls a core BBB gene expression program of over 100 genes involved in tight junction organization\, nutrient and ion transport\, and efflux pump function. To harness this mechanism therapeutically\, we developed\, optimized\, and patented a fully human monoclonal antibody (Anti-Unc5B) that specifically blocks Netrin-1 binding to Unc5B. \nFollowing intravenous injection\, Anti-Unc5B induces a brain-wide and brain-specific\, opening of the BBB for up to six hours\, prior to barrier resealing. This transient permeability window is size-selective\, cumulative and safe. Using this BBB-permeabilizing strategy\, we could enhance the delivery and efficacy of a wide variety of therapeutic molecules including chemotherapies\, tyrosine kinase inhibitors and nanobodies during GBM progression\, resulting in increased tumor cell apoptosis\, reduced tumor growth and improved survival. \nBiosketch \nI completed my PhD in the laboratory of Pr. Andreas Bikfalvi under the supervision of Dr. Clotilde Billottet (INSERM U1029\, France)\, where I identified the chemokine receptor CXCR3-A and its ligand CXCL4L1 as a novel signaling pathway essential for brain tumor progression (Boyé K. et al.\, Nature Communications 2017; Boyé K. et al.\, Scientific Reports 2017; Quemener C et al. Cancer Research 2016). However\, like most therapeutic molecules developed for neurological diseases\, intravenously injected CXCR3-A inhibitors were unlikely to efficiently cross the blood–brain barrier (BBB). \nTo overcome this limitation\, I joined the laboratory of Pr. Anne Eichmann at Yale University (New Haven\, USA) as a postdoctoral fellow (2017-2022)\, where I investigated mechanisms regulating vascular barrier integrity (Genet G.\, Boyé K. et al. Nature Communication 2019; Zhang F. et al. Science 2018)\, with a particular focus on the BBB (Boyé K. et al.\, Nature Communications 2022). I identified the endothelial Unc5B receptor and its ligand Netrin-1 as a key signaling pathway required to maintain BBB integrity. I subsequently developed a human monoclonal antibody that block Netrin-1 binding to Unc5B\, enabling a reversible and controlled BBB opening leading to brain delivery of bioactive molecules (Two Patents in 2023). \n  \nSince 2023\, I am a Group Leader (CRCN) at the Paris Cardiovascular Research Center (INSERM U970). My group is currently funded by an FRM “Amorçage jeunes équipes” (2023-2026)\, an ANR PRC (Coordinator; 2026 – 2028) and an INCa PLBIO (Coordinator; 2026 – 2028). My research program focuses on: \n\nOptimizing brain drug delivery beyond the BBB during brain tumor progression.\nEnhancing BBB integrity in diseases with BBB dysfunction.\n\nWe recently reported a BBB gene-expression program zonated along the arteriovenous axis that can be tuned to increased brain drug delivery (Furtado et al.\, PNAS 2024). \n  \nIn 2024 I co-founded D2B3 (https://d2b3.bio)\, a start-up company spun out of Yale University\, aiming to improve brain drug delivery in patients using our patented BBB-permeabilization technology.

It traces the evolution of cancer theories from cell-based to microenvironment-focused models\, highlighting increased complexity in causal interpretation due to tumor heterogeneity. The “Hallmarks of Cancer” framework is discussed as a descriptive tool that lacks causal clarity\, prompting the use of multi-scale and formal causal models. \nTwo main theoretical approaches—Somatic Mutation Theory and Tissue Organization Field Theory—are compared and integrated\, with probabilistic models illustrating the variable influence of genetics\, cellular environment\, and microenvironmental factors. Case studies on diffuse gliomas and clear cell renal carcinoma demonstrate how advanced causal modeling (using DAGs\, SEM\, and Bayesian approaches) can clarify the interplay among invasion\, immunity\, metabolism\, and genetic mutations. The research also identifies key molecular pathways and therapeutic targets. \nMethodologically\, the work highlights the challenges of validating complex causal networks and advocates for systematically integrating causal analysis in biomedical research. A holistic\, interdisciplinary approach is essential for advancing both fundamental understanding and clinical applications in oncology. Additionally\, it is shown how biomedical research can enhance philosophical conceptual analysis and philosophical concepts biomedical research \nBiosketch \nBorn in Hungary and raised in Germany\, I hold both French and German citizenships. I graduated in medicine in France and completed my residency in hematology and oncology at the University of Kiel (Germany). I then pursued a PhD in science at INSERM in Paris\, where my research focused on the biology of growth factors\, particularly FGFs\, and their role in regulating the phenotype of endothelial cells.  \nIn 1991\, I joined Professor Daniel B. Rifkin’s laboratory in New York\, where I studied the mechanisms of action of different molecular forms of FGFs. Upon returning to France in 1995\, I joined the CNRS and then the University of Bordeaux as a professor of cell and molecular biology\, where I established and led a laboratory dedicated to tumor and vascular biology\, affiliated with INSERM as of 2000. My unit became part of the BRIC INSERM center in Bordeaux in 2022 (Team 1 of BRIC) Since 2025\, I am an emeritus professor. DESCRIPTION:Dr. Bruno Constantin (CNRS Researcher\, 4CS Laboratory\, University of Poitiers) will give a lecture titled « Ion channels and cancer toward the concept of oncochannels ? » Friday\, May 29\, at 2:00 p.m.\, in the BBS lecture hall. \nEnglish presentation.  \nAbstract \nGrowing evidence indicates that ion channels\, particularly calcium channels\, play central roles in cancer biology. They contribute to key hallmarks of cancer\, including proliferation\, survival\, invasion\, metastasis\, and therapeutic resistance. Cancer stem cells\, in particular\, rely on ion channel–dependent signaling to sustain self-renewal and adaptability. These effects often arise from aberrant channel expression or activity\, enabling cancer cells to hijack physiological signaling pathways. This has led to the emergence of the concept of “oncochannels\,” referring to ion channels that actively drive tumor initiation and progression. Beyond improving our understanding of cancer pathophysiology\, this concept identifies ion channels as promising biomarkers and therapeutic targets. \nBiosketch \nWith a Master’s degree in Physiology and Cellular Biology (electrophysiology\, University of Poitiers)\, my research focuses on calcium signaling in physiology and disease. My early work addressed calcium dynamics during myogenesis and actin cytoskeleton remodeling (Ghent University).  \nAt CNRS\, I demonstrated the role of dystrophin in calcium homeostasis and ion channel regulation in skeletal muscle\, leading to studies on TRPC/α-syntrophin complex and their involvement in Duchenne muscular dystrophy\, as well as the therapeutic potential of mini-dystrophin. \nSince 2014\, I directed STIM and 4CS lab. My research has expanded to cancer and stem cell biology and has focused on calcium channels in neural and cancer stem cells\, particularly store-operated calcium entry (SOCE)\, and their roles in glioma cell self-renewal and invasion. I have also contributed to projects on TRP channels in invasive cancer cell migration across various cancer models.

We show that premenopausal women are more susceptible to certain cancers and identify ECAD as a key mediator of estrogen response. In melanoma\, an ECAD–βCAT–ERα–GRPR pathway drives female-specific tumor aggressiveness\, while targeting GRPR or ERα reduces metastasis. DESCRIPTION:Frederic Delom and his 2.2 team have invited Cédric Chaveroux to present his research. \nCédric Chaveroux\, Laboratoire de Biologie tissulaire et Ingénierie Thérapeutique – UMR 5305\, Lyon \nTitre :\nA non-canonical RNA damage signaling drives cell plasticity under translational stress: implications for colorectal tumor cell state transitions. (english presentation). \nBiosketch \nDr Cédric Chaveroux is a CNRS researcher and head of the Microenvironmental Stress Signaling and Epithelial Cell Fate team. His work focuses on the mechanisms by which epithelial tumor cells adapt to stress\, with a particular emphasis on RNA damage responses and cellular plasticity. His research investigates signaling pathways activated in response to alterations in RNA integrity\, particularly under conditions of translational stress and disrupted cellular homeostasis. He is especially interested in GCN2 kinase–mediated signaling as a key sensor and integrator of stress\, capable of reprogramming gene expression and cell fate decisions. In this context\, he explores how translation perturbations induced by microenvironmental and/or therapeutic stress contribute to the remodeling of cellular states and promote tumor progression.     \nAbstract \nBeyond its role in protein synthesis\, mRNA translation constitutes a central hub for stress response integration. Under microenvironmental or therapy-induced stress conditions\, its disruption triggers a coordinated set of molecular pathways collectively referred to as the RNA damage response. The dynamic interplay between these pathways plays a critical role in determining tumor cell fate. Our recent work identifies novel signaling mechanisms that enable tumor cells to tolerate translational stress\, thereby promoting their survival and plasticity in colorectal cancer. Beyond redefining the key molecular events underlying tumor cell plasticity\, pharmacological targeting of these mechanisms represents a promising therapeutic opportunity.

Due to their selectivity and modularity\, peptides serve as ideal platforms for targeting hard-to-reach protein-protein interfaces and for exploiting endogenous cellular degradation mechanisms. In this context\, two main targets were explored: hDM2\, for its E3 ligase activity and its interaction with the tumor suppressor p53\, and furin\, a proprotein convertase involved in tumor progression.   \nIn the first phase\, constrained ligands mimicking the natural p53 helix were developed as peptide-foldamer hybrids incorporating oligourea segments and macrocyclization patterns derived from structures co-crystallized with hDM2. The exploitation of new cyclization opportunities that stabilize the original alpha-helix—notably through the introduction of cysteines and a mercaptoprolin residue—enabled the stiffening of compounds exhibiting high affinity for hDM2/hDMX. Careful sequence tuning also revealed that polycationic segments induced p53-independent cytotoxicity\, whereas macrocyclic peptide-foldamer hybrids with a net neutral charge restored p53’s antitumor activity in p53wt cancer cells. These optimized ligands thus constitute high-affinity\, stable\, and permeable modules suitable for subsequent integration into architectures that utilize hDM2 as a target or E3 ligase in the development of PROTAC-type peptidomimetic degraders.    \nIn a second phase\, a robust synthetic route for furin peptide inhibitors was established\, enabling the development of analogs active against the recombinant enzyme and in cellular environments. These ligands were then used as a basis for exploring targeted degradation strategies\, including hydrophobic tagging\, PROTACs targeting the Cereblon or von Hippel-Lindau E3 ligases\, and LYTACs targeting internalizing receptors. The results highlight the potential of these approaches while underscoring the constraints imposed by furin’s membrane localization and luminal orientation\, which limit the efficacy of certain degradation modalities.   \nTaken together\, these studies demonstrate that peptidomimetic engineering\, combined with macrocyclization strategies\, represents a promising approach for generating new bifunctional tools capable of selectively degrading oncoproteins. These advances open up new possibilities for the development of next-generation degraders targeting both intracellular and extracellular targets\, demonstrating that such architectures can be rationally optimized to ensure high affinity\, increased stability\, and the ability to direct proteins to proteasomal or lysosomal degradation pathways.  \n  \nNotice of Thesis Defense
