DESCRIPTION:Dr Léa Lemaitre \nPost Doc\, Division of Gastroenterology and Hepatology\, Stanford University\, Stanford\, CA\, USA \ninvited by Christophe Grosset\, team 6 MIRCADE\, Methods and Innovations for the Research in Pediatric Cancers. \nTitle: Targeting Macrophage-Mediated Mechanisms of Immune Evasion as adjuvant therapy in TACE-resistant Hepatocellular Carcinoma. \n  \nAbstract: \nA key obstacle to progress in treatment of hepatocellular carcinoma (HCC) is the notorious tenacity of HCC to recur. Transarterial chemoembolization (TACE) is the most common treatment for unresectable HCC\, however\, it fails to provide a durable cure. The crux of the problem lies in residual cancer cells that persist after TACE. Our goal is to understand how these residual cells escape immune detection and survive after TACE in order to develop effective adjuvant therapies.    \nWe used both in vitro and in vivo methods to mimic TACE- resistance by generating doxorubicin resistant- residual HCC models. We performed single-cell RNA sequencing on 3D tumoroids (31\,000 cells) of doxorubicin resistant vs control HCC in coculture with macrophages. In addition\, HCC patient-derived tumor cells and macrophages were used for validation. We developed an immunocompetent mouse model of syngeneic orthotopic allograft model of HCC\, using doxorubicin-resistant or control HCC cell lines to test adjuvant therapies.    \nUsing scRNAseq of 3D tumoroid model\, we show that doxorubicin-resistant cancer cells exhibit a stem-cell-like phenotype with high expression of stemness markers (CK19\, CD44)\, TGFb pathway (TGFBR1) and cytokines driving pro- tumor macrophage polarization (CXCL8\, CCL2). On the other hand\, the macrophages co-cultured with doxorubicin-resistant HCC showed a pro-tumor phenotype with increased PDL1 expression. We validated this finding using patient-derived samples\, showing that doxorubicin resistant HCC tumoroid co- culture increased PDL1 expression on macrophages. Further\, mRNA FISH showed that the CD68+ macrophages expressed higher levels of TGFB1 in TACE-resistant HCC than primary HCC (p=0.0011). Based on our in vitro analysis we identified the TGFB pathway and PDL1 as potential synergistic targets for adjuvant therapy of TACE-resistant HCC. In our mouse model of doxorubicin-resistant orthotopic allografts\, we showed that treatment with combined TgfbrI and anti-Pdl1 antibodies resulted in significantly reduced liver tumor burden (p=0.01). Flow cytometry analysis revealed immune remodeling with increased infiltration of CD8T cells and M1-like macrophage (CD86+/MHC- II+)\, along with depletion of macrophages (PDL1+/CD206+) in the liver of treated doxorubicin-resistant orthotopic allografts.       \nWe generated in vitro 3D models of TACE-resistance in HCC and identified the mechanistic roles of the PDL1+ macrophage-derived TGFβ1 on stem-like cancer cell persistence. We developed a mouse model of doxorubicin-resistant HCC and demonstrated that combined targeting of Pdl1 and Tgfb1 blocks tumor progression and evokes and anti-tumor macrophage and CD8T cell immune response. Thus combined targeting of PDL1 and TGFB1 holds therapeutic potential as adjuvant therapy in TACE to eliminate residual HCC\, reduce recurrence and improve patient outcomes in HCC.   \n  \nBiosketch: \nMy long-term research goals are to improve immunotherapies for cancer treatment. My objective is to decipher the complexity of the immune system in the tumor microenvironment. My academic training as a pharmacist and research scientist provides me with a strong background in pharmacy\, medicine\, clinical research\, immunology\, and oncology.   \nIn my first year of pharmacy residency\, I was involved in clinical trials\, and I developed a novel chromatographic method to detect errors in the administration of intravenous chemotherapy. During my residency\, I developed a strong interest in oncology research leading me to pursue an integrated masters program under the mentorship of Dr. Bettina Couderc. I analyzed the crosstalk between macrophages and mesenchymal stromal cells (MSCs) in chemotherapy-resistant ovarian cancer leading to 3 co-author publications. This research project reinforced my passion for immune-oncology. To further understand the role of MSCs in cancer resistance to therapies\, I pursued PhD training under the supervision of Dr. Bettina Couderc and Jill Corre\, entitled “Phenotypic and functional characterization of cancer-associated MSCs in multiple myeloma”. This work advanced the understanding of the tumor microenvironment in multiple myeloma and led to the publication of two first-authored publications (1\, 2). During my doctoral and postdoctoral training\, I was also involved in another project to decipher mechanisms of resistance to immunotherapy\, the results were published in Immunity with me as the second author (3). I thus gained strong training and expertise in tumor immunology and novel immune monitoring technologies.          \nThree years ago\, I joined the Dhanasekaran Lab to deepen my expertise in immuno-oncology. I have gained proficiency in advanced technologies such as CyTOF\, spatial immunology and organoids. I have published a manuscript discussing macrophage-mediated mechanisms of resistance to immune checkpoint inhibitors (4\, co-author\, Cancer Res\, 2023) and an article identifying novel mechanisms of resistance to therapy in liver cancer (5\, first author\, Nat Cancer\, 2024). These papers serve as a cornerstone for my current application to develop a precision oncology platform for immunotherapy in HCC. However\, 60% of patients fail to respond or relapse early after induction\, and 95% develop immune-related adverse events (irAEs) such as colitis\, which can be treated with infliximab\, a tumour necrosis factor-alpha inhibitor (TNFi). The impact of TNFi on immune and clinical responses remains to be elucidated. Here we provide evidence that TNF blockade can improve the response to the anti-PD-1 and anti-CTLA-4 combination not only in mice but also in patients enrolled in a phase 1b clinical trial (TICIMEL\, NTC03293784). However\, certolizumab\, but not infliximab\, strongly stimulates immune and therapeutic responses to ICI. Mechanistically\, we have shown that the IgG1 Fc fragment of infliximab impairs the ability of TNF blockade to stimulate the efficacy of ICI therapy. When combined with ICI\, certolizumab decreased the proportion of regulatory T cells (Tregs) and exhausted T cells\, while increasing the proportion of effector T cells in tumors as assessed by single-cell RNA sequencing and spectral flow cytometry. Andrieu-Abadie.  \n  \nOverview of research activities: \nOver the last 25 years\, I have been studying the signalling pathways triggered by some members of the TNF receptor superfamily\, including CD40\, Fas/CD95 and TNFR1/CD120a. In this context\, we have highlighted the role of sphingolipids in the modulation of both apoptosis and immune responses (Ségui et al.\, J. Biol. Chem.\, 1999; Ségui et al\, J. Clin. Invest. 2001; Montfort et al.\, J. Immunol. 2009; Lafont et al.\, Cell Death Differ. 2010). More recently\, we have shown that TNF blockade enhances the CD8 T cell-dependent immune response (Bertrand et al.\, Cancer Res. 2015) and overcomes the resistance to anti-PD-1 (Bertrand et al.\, Nat. Commun. 2017) in mouse melanoma models. Mechanistically\, TNF triggers the activation-induced cell death of CD8+ tumour-infiltrating lymphocytes. We have translated our concept into the clinic in advanced melanoma patients\, where we have shown that two triple therapies combining TNF blockers (infliximab or certolizumab) with anti-PD-1 (nivolumab) and anti-CTLA-4 (certolizumab) are safe for patients\, with promising signs of efficacy in the certolizumab cohort (Montfort et al.\, Clin. Cancer Res 2021). Moreover\, Elevated plasma TNF and enriched TNF pathways in T cells are associated with poorer clinical outcome in patients with advanced melanoma\, reinforcing the notion that TNF may dampen the efficacy of immune checkpoint inhibitors (Virazels et al.\, Int J Cancer\, in press).      \n

However\, 60% of patients fail to respond or relapse early after induction\, and 95% develop immune-related adverse events (irAEs) such as colitis\, which can be treated with infliximab\, a tumour necrosis factor-alpha inhibitor (TNFi). The impact of TNFi on immune and clinical responses remains to be elucidated. Here we provide evidence that TNF blockade can improve the response to the anti-PD-1 and anti-CTLA-4 combination not only in mice but also in patients enrolled in a phase 1b clinical trial (TICIMEL\, NTC03293784). However\, certolizumab\, but not infliximab\, strongly stimulates immune and therapeutic responses to ICI. Mechanistically\, we have shown that the IgG1 Fc fragment of infliximab impairs the ability of TNF blockade to stimulate the efficacy of ICI therapy. When combined with ICI\, certolizumab decreased the proportion of regulatory T cells (Tregs) and exhausted T cells\, while increasing the proportion of effector T cells in tumors as assessed by single-cell RNA sequencing and spectral flow cytometry. Andrieu-Abadie.  \n  \nOverview of research activities: \nOver the last 25 years\, I have been studying the signalling pathways triggered by some members of the TNF receptor superfamily\, including CD40\, Fas/CD95 and TNFR1/CD120a. In this context\, we have highlighted the role of sphingolipids in the modulation of both apoptosis and immune responses (Ségui et al.\, J. Biol. Chem.\, 1999; Ségui et al\, J. Clin. Invest. 2001; Montfort et al.\, J. Immunol. 2009; Lafont et al.\, Cell Death Differ. 2010). More recently\, we have shown that TNF blockade enhances the CD8 T cell-dependent immune response (Bertrand et al.\, Cancer Res. 2015) and overcomes the resistance to anti-PD-1 (Bertrand et al.\, Nat. Commun. 2017) in mouse melanoma models. Mechanistically\, TNF triggers the activation-induced cell death of CD8+ tumour-infiltrating lymphocytes. We have translated our concept into the clinic in advanced melanoma patients\, where we have shown that two triple therapies combining TNF blockers (infliximab or certolizumab) with anti-PD-1 (nivolumab) and anti-CTLA-4 (certolizumab) are safe for patients\, with promising signs of efficacy in the certolizumab cohort (Montfort et al.\, Clin. Cancer Res 2021). Moreover\, Elevated plasma TNF and enriched TNF pathways in T cells are associated with poorer clinical outcome in patients with advanced melanoma\, reinforcing the notion that TNF may dampen the efficacy of immune checkpoint inhibitors (Virazels et al.\, Int J Cancer\, in press).      \n

DESCRIPTION:Invited seminar by team 6 MIRCADE (Methods and Innovations for the Research in Pediatric Cancers.) headed by Christophe Grosset. \nRomain PARENT\nHepatitis Viruses & Liver Pathogenesis (PathLiv) Laboratory – UMR 1350\nInserm – Claude Bernard Lyon 1 University – Lyon Hepatology Institute \nThe interest of liver innervation in chronic liver disease and liver cancer. \nAbstract\nHepatocellular carcinoma (HCC) care has long been hampered by the enigmatic nature of disease onset across patients\, evolution\, as well as of response or resistance to treatment. Hepatic neural cells are likely the least studied liver cell type and mediate patients singularities from the ANS to the organ in real-time. These cells\, belonging to the autonomic nervous system (ANS)\, mediate liver/whole body crosstalk. Pathological innervation of the ANS has been identified in cancer\, nurturing tumor stroma and conferring stronger carcinogenic properties.\nIn this presentation focusing on a pathogenic continuum between normal liver and HCC\, we will present evidence for: (i) deep alterations of neural markers along this spectrum\, (ii) the presence of neural pools of cells in lesions\, (iii) the introduction of a novel neuronal score-based approach for stratification with respect to pathological and immunological considerations\, and (iv) functional studies causally demonstrating the interest of these notions for rational and accessible therapeutic intervention.     \n  \nBiography\nI obtained a PharmD at the U. of Aix-Marseille 1 in 2001 and a PhD in hepatic virology in 2004 through a PhD exchange program between the Center for the Study of Hepatitis C at the Rockefeller University of New-York and the Inserm Unit 271 in Lyon\, working on the early HCV tissue culture systems.\nI then spent four years at the Fred Hutchinson Cancer Research Center in Seattle working on HCV virology and cell translation. I obtained an Inserm CR2 position in 2009.\nI have been till then leading an independent group within Fabien Zoulim’s PathLiv laboratory\, gradually evolving from translational virology towards the study of liver innervation in pathology\, through a long term work on the axonal guidance cue netrin-1.\nMy group’s work on netrin-1 have ended up in the Phase III HCC-NET1 clinical trial which has for goal to target the axonal guidance factor netrin-1 in HCC. I recently obtained a Master degree in philosophy. As a teaching unit head at the Oncology Master of Lyon 1\, I also tackle these issues through the mind-body philosophical paradigm with the students.   \n

DESCRIPTION:BRIC Team 3\, Liver Cancers and Tumor Invasion\, co-directed by Violaine Moreau and Frederic Saltel\, is pleased to invite Jean-François Côté\, President and Scientific Director of the Institut de recherches cliniques de Montréal (IRCM)\, Canada\, to present a seminar open to the scientific community.\nJean-François Côté is co-supervisor of Mélissa Correia De Oliveira‘s thesis\, team 3 BRIC. \n\nTitle : Mechanistic Insights into Tumour Growth\, Metastasis\, and Dormancy in Cancer Progression.\n\n\nAbstract\n\nMetastasis is the primary cause of cancer-related deaths\, yet the biological processes that enable tumor cells to spread\, survive in distant organs\, and eventually form secondary tumors remain incompletely understood. Gaining insight into these mechanisms is essential for developing strategies to prevent and treat metastatic disease across cancer types. \nIn the first part of my presentation\, I will discuss tumour dormancy in breast cancer. To uncover regulators of dormancy\, genome-wide CRISPR screens were performed on two breast cancer cell lines: 4T1 (short dormancy) and 4T07 (prolonged dormancy). 4T07 cells uniquely depended on class III PI3K (PIK3C3) and showed elevated mTORC1 activity via peripheral lysosome signaling. Inhibiting PIK3C3 suppressed this activity in both mouse and human models\, reduced metastasis in 4T07\, and eliminated dormant cells in a HER2-driven model. These results highlight PIK3C3-mTORC1 signaling as a potential therapeutic target to prevent metastatic relapse. In the second part of the presentation\, I will focus on RAS family proteins that regulates key signaling pathways involved in cell growth\, differentiation\, and survival. While canonical RAS proteins are well studied\, the broader group of 35 human RAS-related GTPases remains largely unexplored. Using proximity-dependent biotinylation (BioID) in three cell models\, we mapped interaction networks and subcellular localizations across the family. This generated a high-confidence interactome\, revealing thousands of spatially resolved associations. By analyzing nucleotide-state-specific variants\, we identified context-dependent interactions with effectors and regulators. Comparative clustering highlighted conserved and unique features across subfamilies. Integration with DepMap data uncovered essential crosstalk with regulators Rho GTPases and cytoskeletal dynamics in canonical RAS proteins. Research interests encompass cancer biology\, acute and chronic leukemias\, chemotherapy resistance\, molecular and cellular biomarkers\, functional genomics\, proteomics\, metabolomics\, nanobiomaterials\, regenerative medicine\, cell-based therapies\, and the application of bioinformatics to precision medicine.     \nAbstract: \nAcute leukemias\, including Acute Lymphoblastic Leukemia (ALL) and Acute Myeloid Leukemia (AML)\, are aggressive hematologic malignancies with high biological heterogeneity and limited predictive tools for therapeutic response and disease progression. In this context\, omics sciences offer powerful strategies to unravel the molecular complexity underlying these diseases and support the identification of new biomarkers and therapeutic targets.  \nThis seminar will provide an overview of our translational research efforts combining metabolomics\, clinical peptidomics\, and machine learning. We will discuss the conceptual and methodological foundations for applying mass spectrometry-based omics approaches (LC-MS and MALDI-TOF/MS) to plasma samples\, as well as the integration of artificial intelligence algorithms to enhance biomarker discovery\, risk stratification\, and individualized therapeutic guidance.  \nAdditionally\, we present the rationale and design of predictive modeling studies employing clinical and genomic datasets from AML patients. Johanna Ivaska (University of Turku) Dr. Guillaume Jacquemet (Åbo akademi)\, both located in Turku (Finland). \nDocent at the University of Turku\, Field of application: Biophysics of cancer. \n  \nDissecting the intravascular adhesion and extravasation of Pancreatic Ductal AdenoCarcinoma. \n📆 Tuesday\, September 9 11 a.m. \n📌 BBS conference room\, 2 rue Dr Hoffmann Martinot\, Bordeaux \n  \nAbstract. \nSolid tumor progression is a complex succession of events happening in different spaces and times in the body\, ultimately leading to the formation of life-threatening metastases. To colonize distant organs\, cancer cells are exploiting the circulatory networks as natural paths before stopping in small vessels and crossing the endothelial barrier. \nMy current research focuses on the steps of intravascular adhesion and extravasation (crossing of the vessel border) of pancreatic ductal adenocarcinoma (PDAC) cell lines. My main objectives are to decipher the mechanisms allowing the cells to break the endothelial barrier\, in an attempt to identify key factors and actors during the process. I work with microfluidic models to better reproduce the physiologic conditions of perfused vessels\, zebrafish embryos which are transparent and very rapidly develop a complex vascular system and murine models. All three are coupled with quantitative and high-resolution live imaging and to single cell characterization approaches (scRNAseq\,mass cytometry\,…). \nMy ongoing results support the idea that heterogeneity of cancer\, even among laboratory cell lines\, is driving different adhesion/extravasation routes that are dramatically understudied. My talk will give an overview of my postdoc projects. 1. FlowVision\, our deep-learning based image analysis framework\, allowing the dissection of the arrest to stabilisation of circulating PDAC and immune cells (Follain et al. BioRxiv 2024). 2. An unbiased enrichment-based screening for key actors of the intravascular adhesion step. 3. The discovery of a novel extravasation route used by PDAC cells. \n  \nBio. \nI did a PhD in life science in the lab. of Dr. Jacky Goetz (Strasbourg). I studied the role of blood flow forces on arrest\, adhesion and extravasation of circulating breast/mammary cells\, using multiple imaging modalities\, mostly applied to microfluidics and zebrafish embryos. I described endothelial remodeling around arrested tumor cells as an extravasation mechanism of cancer cells (Follain et al. Dev cell 2018). Our works demonstrated that not only size exclusion but also shear forces are biophysic parameters driving the metastatic spreading (Follain et al. NRC 2020).\nI moved to Turku as a shared postdoc between the well established Cell adhesion lab of J. Ivaska\, expert in integrin biology\, and the newly born lab of G. Jacquemet focusing on cellular protrusion and cell migration. I used similar models to study the metastatic cascade of pancreatic ductal adenocarcinoma. I added to my skill set deep-learning based image analysis (Follain et al. BioRxiv 2024)\, imaging techniques\, murine models and more. \nI am now looking for the next step of my career in France..!

This thesis focuses on hepatoblastoma\, the most common malignant liver tumor in children. Based on 3D electron microscopy images\, a digital twin of the tumor tissue was created using innovative tools to automatically segment and identify biological structures such as cells\, nuclei or blood capillaries. This information was then used to analyze the internal organization of the tissue using unsupervised learning methods. She is currently serving as an associate professor at the Department of Molecular Medicine and heading the Stem Cell Biology Laboratory at Universiti Malaya\, Malaysia. Exploring the potential of stem cells in development of therapy for degenerative diseases and targeting cancer stem cells are the primary focus of her research. More details can be found in her page: umexpert.um.edu.my/selvee.html and Web of Science Researcher IDF-3044-2012. \n\nAbstract  \nStem cell plasticity is the key concept in developmental biology\, enabling the zygote and inner cell mass to give rise to all 208 specialized cell types through exquisitely precise signalling and cell–cell communication. I will share on how embryonic stem cells can be used to understand and engineer cell fates\, focusing on my work on neuronal and hepatic differentiation. Both embryonic and adult stem cells offer a versatile reservoir for generating therapeutically relevant cells in regenerative medicine\, this talk will cover the work done in our laboratory on developing regenerative therapies using stem cells in combination with other approaches.  \nIntriguingly\, in oncology\, cancer stem cell plasticity fuels tumour heterogeneity\, therapy resistance\, and relapse\, presenting persistent challenges for effective treatment. In the interest of BRIC researchers\, I will devote most of my talk to how we model cancer stem cells in culture\, using these models to unravel the molecular regulation governing plasticity in malignant contexts\, with a particular focus on hepatocellular carcinoma. Duran\nSpanish National Research Council (CSIC) – Séville\nGroup Leader of the Group of Metabolism and Cell Signaling\n \n“Metabolic imbalance as a therapeutic strategy in cancer.” \n  \nAbstract \nDuring last years\, a great amount of effort has been dedicated to elucidate the crosstalk between cellular metabolic and bioenergetic flows with signaling processes\, and how this interaction contributes to coordinate the growth and homeostasis of cells and tissues. Particularly\, how these interaction mechanisms are deregulated in cancer at the molecular and cellular level is a matter of intense research\, with the aim of using this knowledge to propose new therapeutic alternatives. Our research has established the processes of interaction between the metabolism of the amino acid glutamine\, the most abundant in human blood and the most important from an energetic point of view for cells\, with cell signaling via the mTOR pathway\, a protein complex essential in the regulation of cell growth and metabolism\, and autophagy. More recently\, we focused our research on metabolic adaptation associated to therapy resistance in glioblastoma (GBM)\, one of the tumor types with the poorest survival outcomes. This work resulted in the identification of FGFR1 pathway as a major regulator of the signaling and metabolic rewiring associated with temozolomide (TMZ) resistance in GBM. Hence\, we described a mechanism of resistance that operates at two major levels. First\, a p53-mediated regulation of cell cycle inducing cell cycle arrest to allow DNA repair in response to TMZ. And second\, a complete metabolic rewiring promoting lipid catabolism and preventing lipid peroxidation. Both the p53-mediated response and the metabolic adaptation are controlled by FGFR1\, as inhibition of the FGFR1 pathway completely abolishes this signaling and metabolic reprograming\, restoring sensitivity to TMZ. Our results also indicated a correlation of FGFR1 activity with poor prognosis in GBM patients\, and validated the treatment of TMZ in combination with FGFR1 inhibitors as an efficient strategy to induce tumor cell death in pre-clinical animal models. This data position the receptor FGFR1 as a very promising candidate for evaluation in future clinical approaches to limit the development of therapy resistance to TMZ in GBM patients. \n  \nBiosketch \nI obtained my PhD in Biochemistry at the University of Seville (Spain) in 2005 working on cellular bioenergetics. Then\, I joined the lab of Prof. Eyal Gottlieb at the Beatson Institute for Cancer Research (Glasgow\, UK) as a postdoctoral researcher to extend my expertise on broader aspects of cancer metabolism. Later\, I moved to the lab of Prof. Michael N. Hall at the Biozentrum (Switzerland) as a senior postdoc to study the de-regulation of cellular signaling during cancer. In 2013 I joined the Institut Européen de Chimie et Biologie (Pessac\, France) as a Group Leader (INSERM\, CR position)\, integrating my previous expertise to investigate the crosstalk between metabolism and cell signaling in cancer cells. In 2018 I moved my lab to the Centro Andaluz de Biología Molecular y Medicina Regenerativa – CABIMER\, upon my assignment as Senior Research Associate (Científico Titular) at CSIC. Since them\, I am the Group Leader of the Group of Metabolism and Cell Signaling\, and in the period 2019-2022 I was the Director of the Department of Dynamics and Cell Signaling at CABIMER. In the period 2020-2024 I was the Deputy Director of CABIMER. Currently\, I am a member of the Executive Committee of the CONEXION CANCER-CSIC\, and I serve as Associate Editor of the journal Cancer & Metabolism. Since January 2025 I serve as permanent evaluator (Gestor) of the Spanish Agency of Investigation (AEI)\, in the Biomedicine – Cancer commission.

DESCRIPTION:Marion Bouchecareilh et Samuel Amintas from Team 8 “Genetic Biotherapies and Oncology” (BioGO) invite two speakers from Paris Cité University\, Center for Inflammation Research\, INSERM 1149\, Sinkus Team (From micro to macro in cancer development) for this seminar open to the scientific community. \n👉 Aurélie Beaufrère \nMorphological and molecular heterogeneity of hepatocellular carcinoma. \nAbstract:\nHepatocellular carcinoma is the most common primary malignant liver tumor. This tumor is highly heterogeneous\, both morphologically\, with eight histological subtypes described\, and molecularly\, with distinct transcriptomic groups described that are linked to both genomic alterations and morphological aspects. Some of these characteristics impact both prognosis and response to treatment.   \nBiography:\nCurrently an MCU-PH in the Pathology Department at Beaujon Hospital\, my hospital and research work focuses on liver pathology\, particularly liver tumors. I am developing projects on the morphological and molecular heterogeneity of primary liver tumors\, as well as multimodal artificial intelligence projects integrating pathological\, radiological\, and molecular data.  \n  \n👉 Etienne Becht \nHigh-throughput single-cell quantification of 100s of proteins using conventional flow cytometry and machine learning. \nAbstract: \nTechniques enabling single-cell resolution proteomic analyses are limited in terms of the number of cells analyzed and/or the number of proteins measured. To overcome these limitations\, we have developed “Infinity Flow\,” a method for analyzing flow cytometry data in parallel using machine learning models. By enabling the pseudo-quantification of hundreds of proteins in millions of single cells\, this technique allows for in-depth mapping of the microenvironment of complex tissues. We are currently applying it to the study of liver cirrhosis and hepatocellular carcinoma.    \nBiography: \nAs an engineer by training\, I completed a thesis in cancer immunology under the joint supervision of Hervé Fridman (PUPH) and Aurélien de Reyniès (bioinformatician) at the Cordeliers Research Center in Paris. There\, I developed MCP-counter\, a method for estimating the composition of the tumor microenvironment from the bulk transcriptome. I then completed two postdoctoral fellowships\, first at the Singapore Immunology Network on mass cytometry data analysis\, then in Seattle on parallelized flow cytometry data analysis. I then worked in R&D at Servier (Computational Medicine team) before joining Valérie Paradis’ laboratory at the hospital as an INSERM research fellow. My project focuses on profiling the hepatic microenvironment using parallelized flow cytometry and developing bioinformatics methods for analyzing cytometry data.     \n
