We use organoids as model systems\, heterocellular structures that recapitulate the complexity of in vivo tissue dynamics\, including stem cell differentiation and cellular plasticity. Our research integrates bioenergetics\, metabolomics\, and other omics approaches with cutting-edge 4D live imaging to capture metabolic changes across different scales with high temporal resolution. By genetically introducing fluorescent-based reporters\, we monitor metabolite levels within distinct cellular compartments while simultaneously tracking cell type specification. This long-term live imaging data is analyzed using machine learning-based single-cell tracking coupled with advanced analytical pipelines. In this presentation\, I will discuss our findings on how lactate\, traditionally considered a metabolic waste product of tumors\, plays a regulatory role in cancer stem cell differentiation and plasticity through the epigenetic modulation of oncogenes such as MYC.     \n www.RodriguezColmanLab.org \n

A better understanding of how this therapeutic approach works\, and more particularly how killer lymphocytes access tumors during immunotherapy\, could improve the efficacy of the treatments. We recently discovered the essential role played in this process by specialized blood vessels known as tumor-associated high endothelial venules (TA-HEVs) (Asrir* et al. Cancer Cell 2022). TA-HEVs share important phenotypic and functional characteristics with HEVs in lymph nodes and other lymphoid organs (Moussion and Girard\, Nature 2011 ; Girard et al\, Nature Rev Immunol 2012). For the first time\, we were able to film the lymphocytes infiltrating TA-HEV walls to enter the tumors. We found that increasing the proportion of TA-HEVs in a tumor improves the infiltration of stem-like CD8+ T cells\, the efficacy of the immunotherapy and leads to the eradication of the tumors. Finally\, we showed that the likelihood of recovery of metastatic melanoma patients treated with anti-PD-1 plus anti-CTLA-4 immunotherapy is increased when a large number of TA-HEVs are present in tumors. Nature 479\, 542-546. \n  \nBiosketch \nJean-Philippe Girard (PhD) currently holds a position of Senior Research Director at INSERM (French NIH) at IPBS-CNRS\, a large Research Center from the CNRS and the University of Toulouse (France). He received his Ph.D. in Molecular Biology from the University of Toulouse and did his postdoctoral training in Immunology and Vascular Biology at Harvard Medical School (USA) with Pr Tim Springer. Jean-Philippe Girard’s research focuses on the role of high endothelial venules (HEVs) and interleukin-33 (IL-33) in immunity\, inflammation and cancer. He discovered the alarmin cytokine IL-33 as a nuclear factor of HEV (NF-HEV). HEV blood vessels are portals of entry for lymphocytes into lymphoid organs (Moussion and Girard\, Nature 2011; Girard et al\, Nature Rev Immunol 2012). He showed that blood vessels with HEV characteristics designated tumor-associated HEVs are the main sites of lymphocyte entry into tumors\, and are associated with response to immune checkpoint blockade (Asrir* et al Cancer Cell 2022). Dr. Girard is a Highly Cited Researcher in Immunology according to the Web of Science (HCR Index 2023). He received several Scientific Prizes for his work\, including the CNRS Silver Medal (2103)\, and Prizes from the National Academy of Sciences (2012)\, the National Academy of Medecine (2013)\, ‘Fondation pour la Recherche Médicale’ (2016)\, ‘Fondation de France’ (2018) and ‘Fondation ARC’ (2024).       \nJean-Philippe Girard is invited by Team 9 Sarcotarget to sit on the jury for Florent Peyraud’s thesis defense on the same day.

DESCRIPTION:\n\n          NEURO-ONCOLOGY MEETS NEUROSCIENCE\n         \n\n  \n\n\n          The Bordeaux Cancer Neuroscience (BCN) Initiative\n         \n\nSymposium organized by Andreas Bikfalvi\, Ahmad Sharanek\, Thomas Mathivet\, Lucie Brisson\, Antonio Pagano Zottola équipe 1 de notre unité BRIC\, Valentin Nägerl (Institut interdisciplinaire de neurosciences\, IINS) and Thomas Daubon (Institut de Biochimie et Génétique Cellulaires\, IBGC) \n\nGlioblastoma (GB) is the most aggressive brain tumor characterized by high proliferation rate\, core necrosis\, microvascular proliferation and tumor infiltration. GB treatment consists in tumor resection by surgery and radiotherapy plus concomitant chemotherapy with temozolomide (Stupp et al\, 2005). However\, GB still remains incurable with a low median survival rate of approx. 15 months.    This is due to resistance to treatment mainly caused by the presence of tumor cells that escape tumor resection by invading the normal brain parenchyma. Recent developments in immunotherapy did not succeed in overcoming this challenge. Unfortunately\, the mechanisms underlying GB development are still not fully understood and current model systems addressing these questions are not adequate\, impeding progress for the field. It has been shown that the tumor microenvironment (TME) in GB partially controls tumor growth and invasion\, and constitutes a therapeutic target (Bikfalvi et al\, 2022). Tapas Kundu to present a seminar. \nPr Tapas Kundu \nTranscription and Disease Laboratory\, Molecular Biology and Genetics Unit\, Jawaharlal Nehru Centre for Advanced Scientific Research (JNCASR) – India \n  \nChromatin meets autophagy: A tale of nonhistone chromatin protein mediated genome organization and transcription regulation \nThe eukaryotic genome is intricately organized on a beads-on-string structure into nucleoprotein filaments. These filamentous structures consist of nucleosome beads\, composed of approximately 147 base pairs of DNA and four octameric core histones. The folding pattern of these nucleosomes varies depending on the cellular state\, ensuring the overall stability and balance within the organism. In addition to DNA and histones\, various non-histone chromatin-associated proteins along with RNA molecules also play crucial roles in fine-tuning the organization and function of the genome. Our laboratory discovered that a highly abundant nuclear protein\, human transcription coactivator PC4\, is a non-histone component involved in the genome organization\, stability\, and maintenance of the epigenetic state. By employing atomic force and electron microscopy\, we have demonstrated that PC4 directly interacts with core histones and linker histone H1 thus compacting the genome very efficiently. Our experimental results obtained from ATAC-Seq and RNA-Seq analysis provide valuable insights into the regulatory role of PC4 in genome organization. We observed that when PC4 is absent\, there is a significant increase in genome decompaction accompanied by hyper-acetylation levels and abnormal gene expression patterns. Interestingly\, we discovered that embryonic development was impaired in mice lacking PC4\, indicating its essential role for survival. However\, intriguingly\, cell lines with reduced levels of PC4 exhibited enhanced cell proliferation rates and demonstrated resistance to radiation exposure. Further investigation revealed that the mechanism behind this radiation resistance involves the upregulation of autophagy pathways upon depletion of PC4. Remarkably\, our ATAC-Seq data unveiled pervasive genomic decompaction coupled with heightened acetylation specifically at multiple loci related to autophagy-associated genes within these knockdown cells. A similar occurrence was observed in both brain and spleen-specific knockout models of PC4 in mice as well as C. elegans. However\, it is worth noting that the brain-specific knockout of PC4 did not exhibit a pronounced phenotype apart from noticeable impairments in memory extinction. Interestingly\, when PC4 was absent\, there was a significant increase in the expression level of another non-histone protein that interacts with PC4. He joined JNCASR as in 1999. Prof. Kundu has made significant contributions in the area of regulation of gene expression and its link to disease and therapeutics. He is not only elucidating the mechanisms of transcription regulation through the epigenetic modifications but also targeting them to design new generation diagnostics\, as well as therapeutics. Over the years\, he has published several research papers in many international journals. Several patent applications from the laboratory have been granted and some are under process\, which include several academically important research reagents with potential commercial values\, some of which have already been commercialized by renowned companies. He is a fellow of four major national academies of India and served as an editorial board member of the Journal of Biological Chemistry (JBC) from 2011-2016. Presently\, he is serving as an “Associate Editor” of the Journal of Biochemistry (Oxford University Press)\, Series Editor of Subcellular Biochemistry (Springer Nature)\, and the reviewing Editor of eLife Journal. Under his leadership\, several drug molecules reached Phase-I clinical trials and could be licensed to industries for further development. He also led and completed Phase-III clinical trials for anti-Covid 19 therapy which is awaiting DCGI approval. He was instrumental to establish the ‘Asian Forum for Chromatin and Chromosome Biology and ‘Chemical Biology Society\, India’. Besides fundamental research\, Prof. Kundu is also involved in teaching and organizing science outreach programs. His popular lectures on ‘Genes\, Disease and Therapeutics’ have benefitted high school and college students greatly. \n  \nHe is the recipient of several awards\, noteworthy among which are: the Shanti Swarup Bhatnagar Prize from CSIR (2005)\, the National Academy of Science\, India- Reliance Industries Platinum Jubilee Award (2008)\, the Sir JC Bose National Fellowship from DST (2010)\, the GD Birla award for scientific research (2011)\, The Ranbaxy Research Award 2011 in the field of Medical Sciences – Basic Research and India Innovation Award 2012 given by Merck Millipore 2012 (First place)\, Shri Om Prakash Bhasin Award for the year 2019 in the field of Health  Medical Sciences by the Shri Om Prakash Bhasin Foundation\, New Delhi\, Sir M. Visvesvaraya Lifetime Achievement Award for Senior Scientists / Engineers (2021)\, and ICBS-2022 Global Lectureship Award by International Chemical Biology Society (ICBS). \n

DESCRIPTION:Christine Varon\,team 04 – Helicobacter-associated digestive cancers\, cancer stem cells and therapeutic strategies \, invites Pr Alain Gobert to present a seminar. \nPr Alain Gobert\nResearch Professor of Medicine\, Division of Gastroenterology\, Hepatology and Nutrition\, Center for Mucosal Inflammation and Cancer\, Vanderbilt University Medical Center\nNashville\, TN\, USA \nRole of polyamines and electrophilic stress in gastrointestinal carcinogenesis: Therapeutic implications. \nGastric cancer (GC) is the fourth leading cause of cancer death worldwide. The intestinal type of GC is initiated by the pathogen Helicobacter pylori\, which infects half of the world’s population. The three biogenic polyamines\, putrescine\, spermidine\, and spermine play key functions in a variety of cellular processes and are essential for cellular homeostasis. Using animal models of H. pylori-mediated diseases\, we showed that the enzyme spermine oxidase (SMOX)\, which back-converts spermine to spermidine\, plays a major role in development of gastritis and gastric dysplasia. This occurs notably through the generation of acrolein\, a reactive aldehyde that initiates lipid peroxidation and further generation of bifunctional electrophiles. A potent scavenger of these molecules attenuates gastric carcinogenesis. These models\, which include organoids\, spheroids\, organs-on-a-chip and bioprinted tissues\, are currently undergoing exponential development\, thanks in particular to the coincidence between advances in stem cell biology and technologies enabling the topology and composition of the tissue microenvironment to be controlled. As well as being an alternative to animal models\, these models are of major interest for understanding developmental mechanisms\, disease modeling\, drug screening\, regenerative medicine and personalized medicine. This research is intrinsically interdisciplinary\, requiring the collaboration of biologists\, physicians\, physicists\, chemists and engineers.    \nThe aim of this first edition is to bring together people already using or wishing to develop these models\, to share their current or future projects.
