Due to their selectivity and modularity\, peptides serve as ideal platforms for targeting hard-to-reach protein-protein interfaces and for exploiting endogenous cellular degradation mechanisms. In this context\, two main targets were explored: hDM2\, for its E3 ligase activity and its interaction with the tumor suppressor p53\, and furin\, a proprotein convertase involved in tumor progression.   \nIn the first phase\, constrained ligands mimicking the natural p53 helix were developed as peptide-foldamer hybrids incorporating oligourea segments and macrocyclization patterns derived from structures co-crystallized with hDM2. The exploitation of new cyclization opportunities that stabilize the original alpha-helix—notably through the introduction of cysteines and a mercaptoprolin residue—enabled the stiffening of compounds exhibiting high affinity for hDM2/hDMX. Careful sequence tuning also revealed that polycationic segments induced p53-independent cytotoxicity\, whereas macrocyclic peptide-foldamer hybrids with a net neutral charge restored p53’s antitumor activity in p53wt cancer cells. These optimized ligands thus constitute high-affinity\, stable\, and permeable modules suitable for subsequent integration into architectures that utilize hDM2 as a target or E3 ligase in the development of PROTAC-type peptidomimetic degraders.    \nIn a second phase\, a robust synthetic route for furin peptide inhibitors was established\, enabling the development of analogs active against the recombinant enzyme and in cellular environments. These ligands were then used as a basis for exploring targeted degradation strategies\, including hydrophobic tagging\, PROTACs targeting the Cereblon or von Hippel-Lindau E3 ligases\, and LYTACs targeting internalizing receptors. The results highlight the potential of these approaches while underscoring the constraints imposed by furin’s membrane localization and luminal orientation\, which limit the efficacy of certain degradation modalities.   \nTaken together\, these studies demonstrate that peptidomimetic engineering\, combined with macrocyclization strategies\, represents a promising approach for generating new bifunctional tools capable of selectively degrading oncoproteins. Luco  \, team leader of the Chromatin and RNA Splicing team in the Genome Integrity\, RNA\, and Cancer Unit (UMR3348 / U1368) at the Institut Curie\, to give a seminar open to the scientific community. \nA three-dimensional splicing organization important for cell invasiveness. \nMonday\, July 6\, 2026\, 11:45 a.m.\, BBS Conference Room. \nBiosketch \nDr. Luco’s scientific journey began at the Hospital Clinic of Barcelona\, where she completed her PhD in Jorge Ferrer’s lab\, investigating how histone modifications organize genes within nuclear space in the context of inherited diabetes. She then pursued postdoctoral research at the NIH in Tom Misteli’s lab\, where she contributed to pioneering work revealing that chromatin not only regulates DNA but also plays a key role in RNA splicing. In 2013\, she established her own research group at the Institut de Génétique Humaine in Montpellier\, where she expanded her work on how histone marks shape cell type–specific splicing programs through the development of innovative CRISPR-based epigenetic and RNA editing tools. Now based at the Institut Curie in Paris\, her group is exploring how this chromatin-mediated splicing regulation contributes to cancer cell invasiveness and the early steps of metastasis. \nAbstract \nMetastasis and treatment resistance remain the main causes of cancer-related mortality. A major challenge is to target aggressive cells early enough\, before their plasticity enables adaptation through EMT reprogramming. In this context\, the key issue is not only which genes are expressed\, but also how they are processed into different isoforms. Alternative splicing can rapidly rewire protein function without major transcriptional changes and has emerged as a critical regulatory layer in EMT and cancer progression. Indeed\, splicing signatures often outperform gene-expression patterns in identifying poor-prognosis cancers\, suggesting that isoform regulation may better capture the functional adaptations underlying aggressiveness. Yet it remains unclear how transcriptional and splicing programs are activated so rapidly and in a coordinated manner during EMT. We have previously shown that chromatin is a major regulator of splicing dynamics. Using CRISPR–dCas9 epigenome editing\, we demonstrated that exon-localized chromatin marks can drive EMT-associated splicing switches by modulating recruitment of the splicing factor PTBP1 to pre-mRNA. Using an adapted CRISPR genomic screen and antibody-based proximity labeling approaches\, we now propose that invasive reprogramming depends on a dynamic 3D chromatin organization that coordinates transcriptional and splicing regulation through specialized hubs. Distrubing these hubs could be an innovative strategy to target metastasis at its source.

Les PCs sont impliquées dans de nombreux processus physiologiques dus à la variabilité des substrats\, c’est pourquoi la dérégulation de leur expression ou de leur activité peut mener à diverses pathologies dont le cancer.\nBien que PCSK7 soit la convertase la plus ancienne de la famille des PCs\, son rôle physiologique reste très peu caractérisé. Ce n’est que récemment que PCSK7 a été associée au métabolisme des lipides par la régulation des niveaux de l’apoB et de l’apoA-V sans aucune activité protéolytique. Ce qui classe PCSK7\, comme le dernier membre PCSK9\, comme une PC possédant une fonction non enzymatique. PCSK7 est exprimée de façon ubiquitaire dans l’organisme\, cependant elle est retrouvée enrichie dans les organes lymphoïdes au niveau tissulaire et dans les cellules immunitaires\, plus particulièrement dans les lymphocytes T\, au niveau cellulaire. Malgré ces observations\, le rôle de PCSK7 dans la réponse immunitaire reste largement inconnu.\nAfin de définir un possible rôle de PCSK7 dans l’immunité\, nous nous sommes d’abord intéressés aux banques de données humaines pour étudier l’expression de PCSK7 parmi tous les facteurs responsables de la réponse immunitaire. Ces données ont mis en évidence une corrélation positive entre les niveaux d’expression des ARN codant pour PCSK7 et certains points de contrôle immunitaires\, qui sont des protéines de surface inhibitrices de la fonction des lymphocytes T. Nous avons ainsi étudié les niveaux d’expression des points de contrôle immunitaires au niveau intracellulaires et à la surface cellulaire en modulant l’expression de PCSK7 dans différents modèles in vitro tels qu’une lignée cellulaire d’hépatocytes humains\, des lymphocytes T murins isolés de la rate et des lymphocytes T humains provenant d’échantillons de sang de donneurs. Ainsi\, ces études ont démontré le rôle important de PCSK7 dans la régulation des niveaux des points de contrôle immunitaires.\nLes immunothérapies basées sur l’inhibition des points de contrôles immunitaires ont représenté une réelle avancée dans le traitement du cancer mais celles-ci restent peu efficaces pour une majorité de patients. C’est pourquoi nous avons étudié le possible rôle thérapeutique de l’inhibition de PCSK7 dans la progression tumorale\, seule ou en combinaison avec l’inhibition de PCSK9\, déjà reconnue comme un médiateur important de la réponse immunitaire antitumorale. Ainsi\, la délétion simple de Pcsk7 ou de Pcsk9 chez les souris permet une réduction du nombre de métastases hépatiques du cancer colorectal d’environ 50%\, tandis que la double délétion Pcsk7/Pcsk9 permet une réduction du développement métastatique d’environ 90%. DESCRIPTION:Violaine Moreau and Clotilde Billottet\, from Team 3\, have invited Vincent Hyenne to give a seminar. \nVincent Hyenne is the head of the EXPLORE group at the Tumor Biomechanics lab\, INSERM UMR_S1109\, Centre de Recherche en Biomédecine de Strasbourg. \nThe EXPLORE group is studying the priming of metastatic niches by extracellular vesicles \nTitle : Tumor-derived extracellular vesicles facilitate metastasis onset. \nAbstract \nTumor extracellular vesicles (EVs) promote tumor growth and metastasis by mediating the communication between cancer cells and stromal cells. Yet\, how they are generated and how they spread in the organism to modify the microenvironment in distant organs remains unclear. In the past\, we found that Ral GTPases regulate EVs secretion by maintaining the homeostasis of their compartments of origin\, the multivesicular bodies (MVBs). To explore the molecular and cellular mechanisms through which RalA/B drive exosome secretion\, we characterized their proximal interactome and identified EHD4 as a new Ral partner involved in EV secretion. Using various photonic and electronic imaging approaches\, we propose that Ral-dependent recruitment of EHD4 to MVBs promotes their maturation and movement. In parallel\, we are investigating the function of tumor EVs in the formation of pre-metastatic niches. Combining zebrafish and mouse models\, we show that tumor EVs induce a fast mobilization and activation of neutrophils\, which facilitate metastasis initiation.       \nBiosketch \nVincent loves watching and imaging living (and anesthetized) animals. He did his PhD in Jussieu\, Paris on epithelial polarity in mouse early embryonic development (B. Maro lab). He then moved to IRIC (Labbé lab\, Montreal\, Québec) where he studied the establishment of cell polarity\, and its link with vesicular trafficking in C. elegans. Back in France at the IGBMC (Labouesse lab\, Illkirch)\, he got fascinated by exosomes and extracellular vesicles (EVs) and identified new genes required for their secretion in C. elegans. He joined the Tumor Biomechanics lab\, where he got recruited at CNRS in 2016 to study the secretion\, spreading and function of tumor EVs in metastasis in mice and zebrafish. He is in charge of the fish facility and has to admit that zebrafish\, also not being a nematode\, is pretty cool. Vincent is a board member of the French Society of Extracellular Vesicles (FSEV) and the REVE (Réseau Est des Vésicules Extracellulaires). Vincent loves nothing more than traveling with his family\, except maybe a good book\, or a basketball game\, or a glass of good wine. \n

DESCRIPTION:BRIC is organizing a half-day event dedicated to clinical and translational research on Thursday\, October 1\, 2026\, from 2:00 p.m. to 5:30 p.m.\, in the ground-floor lecture hall of the BBS\, in collaboration with the Bergonié Institute and the Bordeaux University Hospital.  \nThe purpose of this afternoon is to raise awareness of the role of clinical and translational research within our unit.
