DESCRIPTION:Mélanie Moreau\, team 03 – Liver Cancers and Tumor Invasion\, will present her thesis on Friday\, December 8\, 2:30 p.m. in the CGFB amphitheatre\, 146 rue Leo Saignat\, Bordeaux. \nsupervisor: Frédéric Saltel \n  \nInvolvement of invadosomes in tumor invasion through two examples\, Ruvbl2 in molecular organization and DDR2 in angiogenesis \nMetastasis is the main cause of death in cancer patients. Around 90% of all deaths are attributed to tumor invasion. Invadosomes are F-actin structures known to be involved in the metastatic process\, particularly through their function of extracellular matrix degradation. This promotes the migration of cells into the extracellular matrix\, and enables them to cross anatomical barriers such as vessel walls. In this thesis\, I focused on the role of Ruvbl2 in invadosome formation and activity. Ruvbl2 is an AAA+ATPase\, member of the R2TP complex\, which plays a chaperoning role in the formation of big protein complexes and is found enriched in rosette-shaped invadosomes. I have contributed to demonstrating that Ruvbl2 is involved in the process of invadosome formation and\, more specifically\, in the organization of these structures into rosettes\, via the R2TP complex. This complex protects several proteins involved in invadosome formation\, including Fak\, from autophagic degradation. I was also interested in the regulation of angiogenesis via invadosome formation and activity in endothelial cells. I demonstrated that\, in the presence of type I fibrillar collagen\, proteasome interacts with DDR2 receptor and negatively regulates invadosome activity. Proteasome inhibition drastically increases invadosome degradation activity. This inhibition stimulates DDR2 activation\, mainly by Src. These results underline the importance of a balance between Src and proteasome activity to avoid cell death. Finally\, this work provides a better understanding of mechanisms of induction\, formation and regulation of invadosomes. It highlights a set of proteins and functions linked to these invasive structures that could be pharmacologically targeted to inhibit metastasis.     \n  \nKey words : Cancer – Invadosomes – Invasion – Ruvbl2 – R2TP – DDR2

Indeed\, the vast majority of patients present at diagnosis with locally advanced or metastatic disease. Its incidence nearly equaling its mortality rate underscores the incurable nature of this cancer. Despite the progress made in understanding the pathogenesis of PDAC\, and the improvement of treatment regimens\, the survival rate after treatment remains poor. Thus\, basic and clinical research remain the best allies of patients and medical teams to make significant breakthroughs and improve the prognosis of this dreaded disease. \nThe ‘Pancreatic Cancer symposium 2023’ will be the third edition of an international meeting that brings together researchers and clinician-scientists at the forefront of knowledge of pancreatic cancer. It is organized by a voluntary group of French researchers (AFRCP). Considering recent progress\, this event will focus on current and future themes in the field. In particular\, the themes of the following sessions\, including interdisciplinarity potentially leading to new advances\, will be proposed to the international research community: identification and targeting of new tumor vulnerabilities\, exploration of the desmoplastic reaction and the tumor microenvironment\, new modalities and technologies for pancreatic cancer research\, knowledge and understanding of early and metastatic disease. \nGiven the international nature of the congress\, we wish to interact with the European association DICE\, https://digestivecancers.eu/ \, in a final session to make a European inventory of patient involvement in the construction of research.

He was previously a Clinician-Scientist and Professor within the Departments of Radiation Oncology and Medical Biophysics at the University of Toronto and a Senior Scientist at the Princess Margaret Cancer Centre. His primary research interests are in tumour hypoxia\, DNA damage signalling and DNA repair in tumours\, and the genomics of prostate cancer progression and cancer treatment response. He is particularly interested in novel clinical trials that intensify cancer therapy to prostate cancer patients whose tumours harbour aggressive genetic changes and hypoxic sub-regions. He has over 300 published papers and book chapters. He is twice a Canadian Foundation for Innovation (CFI) awardee. DESCRIPTION:Dr Barak Rotblat seminar:”How do cells survive glucose starvation” \nBarak Rotblat\, Ben Gurion University (BGU)\, Israël is invited by Andreas Bikfalvi (team 01) for a webinar on February 14 at 11am. \nAbstract: Cells use glucose as a primary source of energy and have developed ways to detect changes in glucose levels to activate survival mechanisms. One such mechanism is the inhibition of a protein called mTOR\, which is responsible for controlling cell growth. It was previously thought that this inhibition leads to a reduction in protein synthesis and preservation of ATP\, but our research has found that the true mechanism is actually a decrease in the translation of mRNA that codes for ACC1\, an enzyme involved in fatty acid synthesis. This leads to a decrease in fatty acid synthesis\, NADPH consumption\, and ROS levels. This mechanism is conserved in human. mouse and yeast cells and exploited by tumor cells to generate tumors in the brain\, a low-glucose environment. \nProf. Rotblat studies the molecular biology of cancer by examining the role of long non-coding RNAs and regulators of mRNA translation. Among other functions\, long non-coding RNAs (lncRNA) control regulation in cells. Protein content analysis shows that a cancerous cell is very similar to a normal cell from the same tissue; for example\, a cancerous breast cell is more similar to a breast cell than to a cancerous cell in the lung. However\, while the protein profiles of healthy and cancerous cells from a given tissue are very similar\, their lncRNA content is very different. Therefore\, lncRNA represents a promising drug and therapeutic target. Regulating mRNA translation is key in the cellular mechanism by which tumor cells adapt to stress. Prof. Rotblat and his colleagues seek drug targets and inhibitor molecules aimed at processes that are used by cancer cells to adapt to starvation conditions\, such as found in a tumor. \nZoom ID 873 524 180 13

Ten years after its discovery\, the genome editing tool has opened up exciting prospects for medicine. « The applications of CRISPR are too numerous to list extensively »\,notes American Jennifer Doudna\, a professor at the University of California\, Berkeley\, who developed the technique with Frenchwoman Emmanuelle Charpentier\, director of the Max Planck Research Center for Pathogen Science in Berlin. Jennifer Doudna publishes an article in the journal Science reviewing the last ten years of CRISPR-Cas9 technology. But as with any medical innovation\, however exciting it may be\, there are undesirable effects to be feared\, or at least monitored. This is the expertise developed by the Inserm team of Prof. François Moreau-Gaudry and Dr. Aurélie Bedel (U1312 Biotherapies of genetic diseases and cancers) at the University of Bordeaux. « Very quickly\, it was observed that CRISPR was less precise than expected: this is called the ‘off-target’ effect\, i.e. the scissors sometimes hit the wrong target or cause changes in the chromosome\,» explains Prof. Moreau-Gaudry. « This is of concern to us because these are molecular events that are found in cancer »\, explains Aurélie Bedel.
