DESCRIPTION:Séminaire de Robert Bristow : “Evolutionary Trajectory of Aggressive Prostate Cancer: Composite Role of Genomics and Microenvironment” \nRobert Bristow\, Manchester Cancer Research Centre\, will speak on February 28 at 2:00 p.m. in the IBGC conference room\, invited by Majid Khatib (team 2). \n\nRobert Bristow is the Director of Manchester Cancer Research Centre and Director of CRUK Major Centre. He was previously a Clinician-Scientist and Professor within the Departments of Radiation Oncology and Medical Biophysics at the University of Toronto and a Senior Scientist at the Princess Margaret Cancer Centre. His primary research interests are in tumour hypoxia\, DNA damage signalling and DNA repair in tumours\, and the genomics of prostate cancer progression and cancer treatment response. He is particularly interested in novel clinical trials that intensify cancer therapy to prostate cancer patients whose tumours harbour aggressive genetic changes and hypoxic sub-regions. He has over 300 published papers and book chapters. He is twice a Canadian Foundation for Innovation (CFI) awardee. DESCRIPTION:Dr Barak Rotblat seminar:”How do cells survive glucose starvation” \nBarak Rotblat\, Ben Gurion University (BGU)\, Israël is invited by Andreas Bikfalvi (team 01) for a webinar on February 14 at 11am. \nAbstract: Cells use glucose as a primary source of energy and have developed ways to detect changes in glucose levels to activate survival mechanisms. One such mechanism is the inhibition of a protein called mTOR\, which is responsible for controlling cell growth. It was previously thought that this inhibition leads to a reduction in protein synthesis and preservation of ATP\, but our research has found that the true mechanism is actually a decrease in the translation of mRNA that codes for ACC1\, an enzyme involved in fatty acid synthesis. This leads to a decrease in fatty acid synthesis\, NADPH consumption\, and ROS levels. This mechanism is conserved in human. mouse and yeast cells and exploited by tumor cells to generate tumors in the brain\, a low-glucose environment. \nProf. Rotblat studies the molecular biology of cancer by examining the role of long non-coding RNAs and regulators of mRNA translation. Among other functions\, long non-coding RNAs (lncRNA) control regulation in cells. Protein content analysis shows that a cancerous cell is very similar to a normal cell from the same tissue; for example\, a cancerous breast cell is more similar to a breast cell than to a cancerous cell in the lung. However\, while the protein profiles of healthy and cancerous cells from a given tissue are very similar\, their lncRNA content is very different. Therefore\, lncRNA represents a promising drug and therapeutic target. Regulating mRNA translation is key in the cellular mechanism by which tumor cells adapt to stress. Prof. Rotblat and his colleagues seek drug targets and inhibitor molecules aimed at processes that are used by cancer cells to adapt to starvation conditions\, such as found in a tumor. \nZoom ID 873 524 180 13

Ten years after its discovery\, the genome editing tool has opened up exciting prospects for medicine. « The applications of CRISPR are too numerous to list extensively »\,notes American Jennifer Doudna\, a professor at the University of California\, Berkeley\, who developed the technique with Frenchwoman Emmanuelle Charpentier\, director of the Max Planck Research Center for Pathogen Science in Berlin. Jennifer Doudna publishes an article in the journal Science reviewing the last ten years of CRISPR-Cas9 technology. But as with any medical innovation\, however exciting it may be\, there are undesirable effects to be feared\, or at least monitored. DESCRIPTION:We are pleased to announce that on September 29-30\, we will be holding the first conference of our new institute\, the BRIC Kick-off meeting. \nThis first event is intended to celebrate the creation of the BRIC and to initiate a series of future symposiums and other congresses within the BRIC. \nThis kick-off meeting will be organized over a day and a half and will include the presentation of the scientific projects of the BRIC members but also of 4 renowned guests: 3 people from SAB Jean-Ehrland Ricci (C3M\, Université Côte d’Azur\, France)\, Ruth Scherz-­Shouval (Weizmann institute of Science\, Israel)\, Urszula Hibner (IGMM\, France) and Phil Jones (The Wellcome Sanger Institute\, Royaume Uni). Finally\, the second day will close with a round table discussion between patient associations and researchers on the theme of patient/researcher collaboration. \n11 sessions are reserved for presentations by BRIC researchers. We already invite each of the team leaders to communicate to us the person who will represent their team during this meeting via this internal link. DESCRIPTION:Anthony Bouter\, UMR5248 CBMN\, Bordeaux and Frédéric Saltel\, U1312 BRIC\, Bordeaux will speak at the Oncosphere’s interdisciplinary seminars the 7 septembre at 12pm\,conference room at IBGC and videoconference. \n \nFrédéric Saltel\nBRIC INSERM 1312\, Team 03 – Liver Cancers and Tumor Invasion. \n\n\nCancer cells transfer invasive properties through tracks\, a novel collagen-associated extracellular vesicle entity \nMetastasis is the leading cause of cancer-related deaths. During this process\, tumor cells acquire invasive and migratory capacities in order to invade the surrounding tissues. To achieve this\, the tumor microenvironment including the extracellular matrix (ECM) is altered to facilitate cancer cell proliferation and dissemination. Extracellular vesicles (EVs)\, such as exosomes or migrasomes\, are already known to induce pro-tumor features such as migration\, promoting tumor development and metastasis formation. Here we highlight a new type of EVs\, referred as tracks\, released by cancer cells during migration and specifically attached along collagen fibers. These tracks\, identified by discoidin receptor 1 (DDR1) enrichment\, are promoted when cell-ECM interactions are increased\, such as in tumors. We characterized these tracks\, their ultrastructure as well as their molecular composition in terms of proteins and nucleic acids\, showing that they are different from classical EVs known so far. Moreover\, these tracks are very stable structures and can be internalized by surrounding cells. After internalization\, they modify the differentiation status and the phenotype of recipient cells\, promoting epithelial to mesenchymal transition\, matrix degradation and invasion. \nThus\, we identified a new class of collagen-associated EVs\, tracks\, that play a role in cell-cell communication by transferring invasive properties.
