SUMMARY:BRIC prestige seminar – Lionel LARUE
DESCRIPTION:As part of the prestigious seminars organized by our BRIC unit\, we are pleased to welcome Dr. Lionel Larue\, Inserm Research Director Emeritus\, and Dr. Véronique Delmas (CNRS Research Director) from the Chemistry and Modeling for Cancer Biology team (UMR9187 / U1196) at the Institut Curie. \nWe will take advantage of their visit to organize meetings with the researchers and students in our unit (see internal email) \nTheir research focused on the role of E-cadherin and β-catenin\, as well as other cadherins (such as N- and R-cadherin)\, in embryonic development\, using sophisticated genetically modified mouse models (knockout/knock-in\, etc.)\, contributing to the study of the role of these proteins and their alterations in pathophysiology. \nThey have long been interested in the development of various tissues and cell types\, including the mammary gland and melanocytes\, and their team has made a name for itself particularly in the study of melanoma. \n  \nThe seminar will be presented by Lionel Larue at 9:30 a.m. on Tuesday\, June 16\, 2026\, in a hybrid format \nBBS conference room\, 2 rue Dr Hoffmann Martinot. location map \nZoom ID 871 6813 2700 \nThe presentation will be in English. \n“The CDH1-ESR1-GRPR axis defines a sex-specific metastatic pathways in cancer ” \nAbstract: \nSex differences in cancer remain underexplored\, limiting precision medicine. We show that premenopausal women are more susceptible to certain cancers and identify ECAD as a key mediator of estrogen response. In melanoma\, an ECAD–βCAT–ERα–GRPR pathway drives female-specific tumor aggressiveness\, while targeting GRPR or ERα reduces metastasis. These findings highlight hormone-dependent mechanisms in non-classical cancers and support sex-specific therapeutic strategies\, including GPCR targeting. \n If scientists from outside the BRIC unit would like to meet with Dr. Lionel Larue and Dr. Véronique Delmas\, please contact David Cappellen at david.cappellen@u-bordeaux.fr. \n  \nLionel Larue CV\nJoin the webinar on Zoom\n  \nCurie Institute report on the article published in Nature in June 2025: Targeting GRPR for sex hormone-dependent cancer after loss of E-cadherin

SUMMARY:Invited seminar - Cédric Chaveroux - 22-06-2026
DESCRIPTION:Frederic Delom and his 2.2 team have invited Cédric Chaveroux to present his research. \nCédric Chaveroux\, Laboratoire de Biologie tissulaire et Ingénierie Thérapeutique – UMR 5305\, Lyon \nTitre :\nA non-canonical RNA damage signaling drives cell plasticity under translational stress: implications for colorectal tumor cell state transitions. (english presentation). \nBiosketch \nDr Cédric Chaveroux is a CNRS researcher and head of the Microenvironmental Stress Signaling and Epithelial Cell Fate team. His work focuses on the mechanisms by which epithelial tumor cells adapt to stress\, with a particular emphasis on RNA damage responses and cellular plasticity. His research investigates signaling pathways activated in response to alterations in RNA integrity\, particularly under conditions of translational stress and disrupted cellular homeostasis. He is especially interested in GCN2 kinase–mediated signaling as a key sensor and integrator of stress\, capable of reprogramming gene expression and cell fate decisions. In this context\, he explores how translation perturbations induced by microenvironmental and/or therapeutic stress contribute to the remodeling of cellular states and promote tumor progression. \nAbstract \nBeyond its role in protein synthesis\, mRNA translation constitutes a central hub for stress response integration. Under microenvironmental or therapy-induced stress conditions\, its disruption triggers a coordinated set of molecular pathways collectively referred to as the RNA damage response. The dynamic interplay between these pathways plays a critical role in determining tumor cell fate. Our recent work identifies novel signaling mechanisms that enable tumor cells to tolerate translational stress\, thereby promoting their survival and plasticity in colorectal cancer. Beyond redefining the key molecular events underlying tumor cell plasticity\, pharmacological targeting of these mechanisms represents a promising therapeutic opportunity.

SUMMARY:Thesis Defense by Mathieu Bourgeais\, Team 2 BRIC & CBMN
DESCRIPTION:Organic Chemistry Thesis Defense: Mathieu Bourgeais\, team 2.1 \nThesis Title: \nSynthesis and Evaluation of Peptide Compounds for the Targeted Degradation of Oncoproteins (french presentation) \nThesis advisors: \nAbdel-Majid Khatib (BRIC team 2.1) et Gilles Guichard (IECB\, CBMN\, UMR 5248) \nDate and time of the defense: \nJune 25\, 2026\, 9 a.m. \n  \nLocation: \nConference room IECB\, 2 Rue Robert Escarpit\, 33600 Pessac \nVideo conference link: \nhttps://sanofi.zoom.us/j/99771203256?pwd=mNK7Iih0btybyAtw0oALnn3dA91D49.1&from=addon \nAbstract: \nThis thesis focuses on the design\, synthesis\, and evaluation of innovative peptide and peptidomimetic compounds intended for the modulation and targeted degradation of oncoproteins. Due to their selectivity and modularity\, peptides serve as ideal platforms for targeting hard-to-reach protein-protein interfaces and for exploiting endogenous cellular degradation mechanisms. In this context\, two main targets were explored: hDM2\, for its E3 ligase activity and its interaction with the tumor suppressor p53\, and furin\, a proprotein convertase involved in tumor progression. \nIn the first phase\, constrained ligands mimicking the natural p53 helix were developed as peptide-foldamer hybrids incorporating oligourea segments and macrocyclization patterns derived from structures co-crystallized with hDM2. The exploitation of new cyclization opportunities that stabilize the original alpha-helix—notably through the introduction of cysteines and a mercaptoprolin residue—enabled the stiffening of compounds exhibiting high affinity for hDM2/hDMX. Careful sequence tuning also revealed that polycationic segments induced p53-independent cytotoxicity\, whereas macrocyclic peptide-foldamer hybrids with a net neutral charge restored p53’s antitumor activity in p53wt cancer cells. These optimized ligands thus constitute high-affinity\, stable\, and permeable modules suitable for subsequent integration into architectures that utilize hDM2 as a target or E3 ligase in the development of PROTAC-type peptidomimetic degraders. \nIn a second phase\, a robust synthetic route for furin peptide inhibitors was established\, enabling the development of analogs active against the recombinant enzyme and in cellular environments. These ligands were then used as a basis for exploring targeted degradation strategies\, including hydrophobic tagging\, PROTACs targeting the Cereblon or von Hippel-Lindau E3 ligases\, and LYTACs targeting internalizing receptors. The results highlight the potential of these approaches while underscoring the constraints imposed by furin’s membrane localization and luminal orientation\, which limit the efficacy of certain degradation modalities. \nTaken together\, these studies demonstrate that peptidomimetic engineering\, combined with macrocyclization strategies\, represents a promising approach for generating new bifunctional tools capable of selectively degrading oncoproteins. These advances open up new possibilities for the development of next-generation degraders targeting both intracellular and extracellular targets\, demonstrating that such architectures can be rationally optimized to ensure high affinity\, increased stability\, and the ability to direct proteins to proteasomal or lysosomal degradation pathways. \n  \nNotice of Thesis Defense
