The intestinal type of GC is initiated by the pathogen <em>Helicobacter pylori<\/em>, which infects half of the world\u2019s population. The three biogenic polyamines, putrescine, spermidine, and spermine play key functions in a variety of cellular processes and are essential for cellular homeostasis. Using animal models of <em>H. pylori<\/em>-mediated diseases, we showed that the enzyme spermine oxidase (SMOX), which back-converts spermine to spermidine, plays a major role in development of gastritis and gastric dysplasia. This occurs notably through the generation of acrolein, a reactive aldehyde that initiates lipid peroxidation and further generation of bifunctional electrophiles. A potent scavenger of these molecules attenuates gastric carcinogenesis.
