DESCRIPTION:Séminaire invité par l’équipe 6 MIRCADE (Méthodes et Innovations pour la Recherche sur les Cancers de l’Enfant) dirigée par Christophe Grosset. \nRomain PARENT\nHepatitis Viruses & Liver Pathogenesis (PathLiv) Laboratory – UMR 1350\nInserm – Claude Bernard Lyon 1 University – Lyon Hepatology Institute \nThe interest of liver innervation in chronic liver disease and liver cancer. \nRésumé\nHepatocellular carcinoma (HCC) care has long been hampered by the enigmatic nature of disease onset across patients\, evolution\, as well as of response or resistance to treatment. Hepatic neural cells are likely the least studied liver cell type and mediate patients singularities from the ANS to the organ in real-time. These cells\, belonging to the autonomic nervous system (ANS)\, mediate liver/whole body crosstalk. Pathological innervation of the ANS has been identified in cancer\, nurturing tumor stroma and conferring stronger carcinogenic properties.\nIn this presentation focusing on a pathogenic continuum between normal liver and HCC\, we will present evidence for: (i) deep alterations of neural markers along this spectrum\, (ii) the presence of neural pools of cells in lesions\, (iii) the introduction of a novel neuronal score-based approach for stratification with respect to pathological and immunological considerations\, and (iv) functional studies causally demonstrating the interest of these notions for rational and accessible therapeutic intervention. \n  \nParcours\nI obtained a PharmD at the U. of Aix-Marseille 1 in 2001 and a PhD in hepatic virology in 2004 through a PhD exchange program between the Center for the Study of Hepatitis C at the Rockefeller University of New-York and the Inserm Unit 271 in Lyon\, working on the early HCV tissue culture systems.\nI then spent four years at the Fred Hutchinson Cancer Research Center in Seattle working on HCV virology and cell translation. I obtained an Inserm CR2 position in 2009.\nI have been till then leading an independent group within Fabien Zoulim’s PathLiv laboratory\, gradually evolving from translational virology towards the study of liver innervation in pathology\, through a long term work on the axonal guidance cue netrin-1.\nMy group’s work on netrin-1 have ended up in the Phase III HCC-NET1 clinical trial which has for goal to target the axonal guidance factor netrin-1 in HCC. I recently obtained a Master degree in philosophy. As a teaching unit head at the Oncology Master of Lyon 1\, I also tackle these issues through the mind-body philosophical paradigm with the students. \n

DESCRIPTION:L’équipe 3 de BRIC\, Cancers du Foie et Invasion Tumorale\, co-dirigée par Violaine Moreau et Frederic Saltel est heureuse d’inviter Jean-François Côté\, président et directeur scientifique de l’Institut de recherches cliniques de Montréal (IRCM)\, Canada\, à présenter un séminaire ouvert à la communauté scientifique.\nJean-François Côté est co-directeur de thèse de Mélissa Correia De Oliveira\, équipe 3 BRIC. \n\nTitre : Mechanistic Insights into Tumour Growth\, Metastasis\, and Dormancy in Cancer Progression.\n\n\nAbstract\n\nMetastasis is the primary cause of cancer-related deaths\, yet the biological processes that enable tumor cells to spread\, survive in distant organs\, and eventually form secondary tumors remain incompletely understood. Gaining insight into these mechanisms is essential for developing strategies to prevent and treat metastatic disease across cancer types.\nIn the first part of my presentation\, I will discuss tumour dormancy in breast cancer. To uncover regulators of dormancy\, genome-wide CRISPR screens were performed on two breast cancer cell lines: 4T1 (short dormancy) and 4T07 (prolonged dormancy). 4T07 cells uniquely depended on class III PI3K (PIK3C3) and showed elevated mTORC1 activity via peripheral lysosome signaling. Inhibiting PIK3C3 suppressed this activity in both mouse and human models\, reduced metastasis in 4T07\, and eliminated dormant cells in a HER2-driven model. These results highlight PIK3C3-mTORC1 signaling as a potential therapeutic target to prevent metastatic relapse. In the second part of the presentation\, I will focus on RAS family proteins that regulates key signaling pathways involved in cell growth\, differentiation\, and survival. While canonical RAS proteins are well studied\, the broader group of 35 human RAS-related GTPases remains largely unexplored. Using proximity-dependent biotinylation (BioID) in three cell models\, we mapped interaction networks and subcellular localizations across the family. This generated a high-confidence interactome\, revealing thousands of spatially resolved associations. By analyzing nucleotide-state-specific variants\, we identified context-dependent interactions with effectors and regulators. Comparative clustering highlighted conserved and unique features across subfamilies. Integration with DepMap data uncovered essential crosstalk with regulators Rho GTPases and cytoskeletal dynamics in canonical RAS proteins.
