DESCRIPTION:Sigve Nakken\nInstitute for Cancer Research at Oslo University Hospital\, Norvège \n📆 4 juin à 14h. \n📌 IBGC (Carreire 146 rue Leo Saignat\, Bordeaux) \nSéminaire organisé par le Département Sciences Biologiques et Médicales (SBM) de l’Université de Bordeaux. \n  \nA suite of computational frameworks to support precision cancer medicine \nCancer precision medicine is an approach to cancer treatment that tailors medical care to the individual characteristics of each patient and their tumor. A cornerstone of this approach is the use of high-throughput genomic profiling technologies for a deep characterization of the molecular features of tumor samples. The diversity and complexity of molecular read-outs\, combined with a rapidly evolving landscape of clinically actionable targets\, poses significant requirements for advanced data interpretation tools. In this talk\, I will present molecular data interpretation frameworks that we are actively developing to address the N-of-1 analytical challenge in precision oncology\, that is to highlight treatment opportunities for each complex cancer case in light of existing knowledge and large-scale reference datasets. Herein\, I will describe challenges and solutions with respect to molecular data integration and harmonization\, the adoption of standards for evaluating the clinical significance of genomic aberrations\, as well as the identification of relevant biomarker-guided clinical trials. \n  \nBio \nSigve Nakken works as a scientist at the Institute for Cancer Research at Oslo University Hospital\, Norway\, with affiliation to the Centre for Cancer Cell Reprogramming (CanCell)\, a Centre of Excellence at the University of Oslo. In addition\, he holds an Adjunct Professor position at the Department of Informatics\, University of Oslo. Nakken’s main interests are tool development for clinical interpretation of cancer genomes\, mechanisms and impact of DNA variation in cancer (soma and germline)\, and knowledge integration for precision cancer medicine. He has a background from computer science (MSc) and computational biology (PhD)\, and extensive experience with analysis of data from high-throughput sequencing technologies. He is the lead developer of multiple bioinformatics tools used in high-throughput cancer biology.

DESCRIPTION:François Moisan\, chercheur équipe 05 – Recherche translationnelle en cancérologie cutanée et maladies cutanées rares (TRIO2) soutiendra son HDR (habilitation à Diriger la Recherche) mercredi 5 juin à 14h\, amphi BBS. \nSujet : Mécanismes cellulaires et moléculaires de la réponse antitumorale au propranolol dans les tumeurs vasculaires. \nFrançois Moisan travaille sur les tumeurs vasculaires cutanées et leur thérapie. La découverte fortuite de l’efficacité du propranolol sur les hémangiomes infantiles (HI) dans l’équipe en 2008 l’a amené lors de son retour des USA en 2013 à travailler sur ce mécanisme d’action. Ce travail à permis de révéler que (1) l’HI est une tumeur riche en noradrénaline (NA)\, un des ligands naturels de la voie béta-adrénergique\, cible attendue du traitement. (2) Les cellules composant la tumeur (cellules endothéliales et péricytes) sont capables de produire et sécréter la NA. (3) L’inhibition par compétition de la NA par le propranolol permet d’inhiber ses effets pro-angiogéniques in vitro. Il a également pu mettre en évidence (4) un troisième type cellulaire déterminant dans la pathogenèse et la réponse au propranolol : le télocyte. Cette cellule stromale impliquée dans la communication cellulaire\, l’architecture et le remodelage tissulaire\, y compris dans la peau se révèle ici (5) comme surexprimant l’aquaporine-1 modulée par le propranolol. Le développement de modèles in vitro de coculture pertinent pour cette étude permet d’explorer les mécanismes cellulaires et moléculaires impliqués dans la croissance et la régression des hémangiomes\, dans le but d’identifier de nouvelles voies thérapeutiques pour les tumeurs et anomalies vasculaires. Le groupe qu’il anime au sein de l’équipe oncodermatologie (TRIO2) s’intéresse également aux angiomes associés aux mutations de GNAQ et aux cancers de types angiosarcomes. \n  \nAvis de soutenance à ce lien \n

During this presentation I will feature published and unpublished data indicating how transcriptional and epigenetic identities are conserved throughout cell division and differentiation.\nMitotic entry correlates with the condensation of the chromosomes\, changes in histone modifications\, exclusion of transcription factors from DNA\, and the broad downregulation of transcription. We found that preventing one chromosome to condense during mitosis causes it to fail resetting transcription. Rather\, in the following interphase\, the affected chromosome contains unusually high levels of the transcription machinery\, resulting in abnormally high gene expression levels\, including various transcription factors. This subsequently causes the activation of inducible transcriptional programs in trans\, even in the absence of the relevant stimuli. Thus\, mitotic chromosome condensation exerts stringent control on interphase gene expression to ensure the maintenance of cell identity across cell divisions (1).\nWe used adipogenesis as a model to find that SUMOylation of chromatin-bound proteins ensures the precision of gene expression and transcription factor dynamics during differentiation (2). However\, the molecular mechanisms by which SUMOylation orchestrates the rewiring of transcriptional programs during initiation of differentiation remains to be established. Our unpublished data indicate that transient\, pharmacological inhibition of SUMOylation in human preadipocytes is sufficient to prime and activate specific enhancers and transcription factors. This causes irreversible hypertranscription of adipogenic genes\, leading to excessive fat accumulation. Given that epigenetic and transcriptional regulators are major SUMO targets\, we propose that SUMOylation maintains a chromatin landscape in undifferentiated cells\, which is critical to preserving the epigenetic fate of mature adipocytes\, protecting against unrestricted adipogenic transcription. This implies that the inherent adipogenic capacity of human adipose stem cells may be tied to SUMOylation and suggests an epigenetic basis for the inter-individual variability in fat accumulation.\nUsing adipogenesis as a paradigm to uncover new molecular pathways by which SUMOylation sustains epigenetic fates\, we expect to contribute to a broader understanding of how transitory disruptions in epigenetic regulation induce stable homeostatic shifts and lead to disease\, like obesity and cancer (3). \n\nRamos-Alonso\, L. et al. My research in the group of Jean-Marc Egly\, at the IGBMC\, was focused on revealing the origin of gene expression defects in patients with rare genetic disorders like Xeroderma pigmentosum (1).\nI then moved to Norway at the Dept. of Microbiology at Oslo Univ. Hospital. For the next eight years\, I continued my research in the group of JM. Enserink on gene regulation in the model organism Saccharomyces cerevisiae. The first four years were dedicated to revealing mechanisms that synchronize cell-cycle progression and gene expression (2). In the next four years I strated investigating the mechanisms regulated by the Small Ubiquitin-like Modifier SUMO at the chromatin level during adaptive response to stress (3) and started exploring the perpetuation of epigenetic and transcriptional memory throw-out mitosis (4).\nNext\, I started my group at Oslo Univ. Hospital and was appointed Associate Professor at the University of Oslo\, where I have been working since 2020. Regroupant les organoïdes\, les sphéroïdes\, les organes sur puce et les tissus bio-imprimés\, ces modèles connaissent actuellement un développement exponentiel\, du fait notamment de la coïncidence entre les avancées dans la biologie des cellules souches et les technologies permettant de maîtriser la topologie et la composition du micro-environnement tissulaire. En plus d’être une alternative aux modèles animaux\, ces modèles présentent un intérêt majeur pour la compréhension des mécanismes de développement\, la modélisation de maladies\, le criblage de médicaments\, la médecine régénérative et la médecine personnalisée. Il s’agit d’une recherche intrinsèquement interdisciplinaire\, nécessitant la collaboration de biologistes\, médecins\, physicien·nes\, chimistes et ingénieur·es. \nLe but de cette première édition est de réunir les personnes utilisant déjà ou souhaitant développer ces modèles afin de partager leurs projets en cours ou à venir. Cette journée permettra aux différent·es protagonistes locaux d’interagir entre eux\, de mutualiser certains efforts\, de promouvoir les échanges et les collaborations. \nDétails pratiques \nLa journée sera organisée autour des sessions thématiques avec une communication invitée et des communications courtes (8 min + 2min de questions) par session. Les pauses café et déjeuner seront accompagnées de sessions « posters ».
