DESCRIPTION:Christine Varon\, équipe 4 – Cancers digestifs associés à l’infection par Helicobacter\, cellules souches cancéreuses et stratégies thérapeutiques\, invite Pr Alain Gobert a présenter un séminaire. \nPr Alain Gobert\nResearch Professor of Medicine\, Division of Gastroenterology\, Hepatology and Nutrition\, Center for Mucosal Inflammation and Cancer\, Vanderbilt University Medical Center\nNashville\, TN\, USA \nRole of polyamines and electrophilic stress in gastrointestinal carcinogenesis: Therapeutic implications. \nGastric cancer (GC) is the fourth leading cause of cancer death worldwide. The intestinal type of GC is initiated by the pathogen Helicobacter pylori\, which infects half of the world’s population. The three biogenic polyamines\, putrescine\, spermidine\, and spermine play key functions in a variety of cellular processes and are essential for cellular homeostasis. Using animal models of H. pylori-mediated diseases\, we showed that the enzyme spermine oxidase (SMOX)\, which back-converts spermine to spermidine\, plays a major role in development of gastritis and gastric dysplasia. This occurs notably through the generation of acrolein\, a reactive aldehyde that initiates lipid peroxidation and further generation of bifunctional electrophiles. A potent scavenger of these molecules attenuates gastric carcinogenesis.
